    The video is great! Thank you very much! I assume it is in fact an in vivo method not in vitro. Otherwise, you wouldn't have needed all this yeast cell machinery to perform the assay.

    good video, I do have a question though. Why do GAL AD and GAL BD bind, when two protein interact? Coulnt the active sites interacting with eath other from the bait and the fish protein be on the opposite sides of those of the GAL proteins, which would mean that the protein interact but the GAL proteins are to far from each other to interact and activate the reporter gene? Or on the other hand isnt it possible as well to have two proteins which dont interact but the the GAL proteins still are able to bind and produce the reporter genes? Shoundt there be a large number of false postive and false negatives with this method?

    Transformation means delivering gene into a cell .....it is possible to do it on bacteria ,yeast or mammalian cell culture. So generally for yeast electroporation is the mode of gene delevery

    Depends what you call in vivo. Some people belive in vivo is tissue level stuff. For Some it is cell level stuff.

    Just because it is happening in an yeast it's not in vivo...the terms are used in pretty loose manner . ( for example cell culture is also considered as in vitro models )

    ​@@anaghamenon7857 Literally, what he has explained to you is not correct. Cell culture is considered to be in vitro because such method is only used to produce biological compounds such as antibodies, instead of to detect a reaction or interaction inside the cells. The majority of the experiment is performed outside of the cells. Yeast 2 hybrid system is an in vivo technique because not only all the proteins are expressed and matured inside the cells, but also the interaction. So, what you have heard before is correct!

    Its a controversy ! One school of thought suggests its in vivo, because the interaction is tested inside an organism . But current view conflicts that. Any way for undergrad textbook level, lets consider it in vivo.
