Hey thanks a lot for this brilliant video but I have a doubt here.... we take 2 components of the transcription factor AD and DBD and bind the 2 proteins one to each.......how can we say the function of the 2 components does not get altered when one of the proteins is attached to it? Pl help me understand

Good question! So the main criteria is that you have to use a strong transcrption factor which initiates transcription of your reporter gene. In that case here they used Gal4p transcription factor: link.springer.com/protocol/10.1385/1-59259-762-9:247
wow that's a very elegant system. Does it mean then that the proteins need to be taken up into the nucleus in order to be tested, is there limitations in terms of size and charge?

Every other source i found doesn't mention anything about Gal4AD recruiting the polymerase, they just mention that transcription happens. Do you happen to have any sources that show that GAL4AD does indeed recruit the polymerase?

Matevz Kosutnik thanks for your question 1. you synthesize a DNA fragment where both sequences are connected and encode a protein with is ´chimericˋ meaning a novel protein out of two individual proteins which are linked via peptide bonds. 2. you can test whether protein A is interacting with protein B (if so, you generate a reporter gene expression)
