  very unlikely the amplified DNA molecules would not be detected because the sybr green dye binds to dsDNA pretty strongly

  to clarify the doubts most people have in the comment section, the first set of primers are added along with the dntps ,buffer , taq polymerase first , then after the first round of amplification, the product from the first PCR reaction is used as the template for the second reaction, where the second set of primers are added.

  Thank you. But 3’ OH group of sugar moiety reacts with 5’ position of phosphate of another sugar moiety rather than 2’ position. Therefore phosphodiester bond forms between 5’ and 3’. So, please change your dna backbone slide
