Determination of Ash Content (Total Minerals)_A Complete Procedure (AOAC 942.05)

Determination of Ash is one of the important proximate analysis for food, feed, vegetable and many other samples. It represents a part of quality of a sample. This method removes all kind of organic matters from a sample which may interfere in other chemical analysis, for example: analysis of heavy metals, minerals.
Moreover, this video represents a complete procedure for determination of Ash from a food sample which will be very helpful for a laboratory analyst.

    Awesome video! Question, I understand combustion reduces food to inorganic mineral ash. I’m confused, if the body uses minerals however the minerals become inorganic after digestion, how is it assimilated?

    Spinoza Black, thanks for your question. Inside a human body, digestive juice creates acidic condition in the stomach to digest you food to convert it into difderent micronutrients and thus releasing minerals. These are further absorbed into the blood vessels.

    Thank for the nice video. I have some questions. My samples are not turning to grey color. I keep it 48 h is the muffle (600 oC) but it still has a light green and reddish color. The ash content is about 30 %. I think it may be too high. My samples are the herb extracts with different concentration of ethanol. I don't know what wrong with my sample or procedure. Hopefully, you see my comment and kindly give me some advice.

    Keep burning for 5 hours. Few samples are remained green and reddish after ashing. Just ensure that the black color is removed completely.

    Fire insident will occure if you open the furnace at a temperature higher than 250 degree celsius.

    Interesting. This was informative. I have a question, hope you see this and reply. So what if I want to get the ash content from meat e.g beef? Do I blend it to homogenize? If I place in in a furnace, would it burnnnnnn, like burn burn that it sticks to the inside wall of that bowl?

    Jalal Shah, Thank you fot your comment. Ash% totally depends on the minerals & heavy metals content present in your honey sample. So high content of heavy metals & minerals means high Ash percent. You can reduce Moisture from your sample by increasing the density of your sample. This can be accomplished by drying the sample at elevated temperature.

    I got your point. Dry 10g of your sample in hot air oven at the same condition as described in Moisture content determination video and cool in desiccator. Then analyse the dried & cooled sample for ash.

    I have a doubt regarding the closing of the crucible while heating. If the lid is closed will burning or combustion takes place properly? I think it has to be heated with the lid open.

    No. The crucible must keep open while heating, according to AOAC method. When you will apply heat at above 550 degree Celsius, the sample will burn out, no matter the crucible is open or close. Crucible is kept closed to avoid loss of ash during burning.

    Mia, In case of plastic granules you can use crucible without lid to determine the ash content as plastic content won't loss during incineration. But in this case, large size of crucible is more preferable to use to avoid sample loss.

    No need. But you can incinerate your sample above 150 degree Celsius for 2 hour after taking sample weight and before going for the ashing into furnace

    We could not provide Ash test documents. Plz follow this video for Ash test of chocolate. Crucible with lid is better than the crucible without lid. But you can use crucible without lid. An Ideal furnace will take only 2 min to reach 250 degree Celsius.

    to determine the ash content of spice powders and wheat flour, semolina is it necessary to do charring or can we directly put the crucible inside the muffle furnace without doing charring?....please help!

    Charring is better because it facilitate to become your sample carbon free very fast. You can analyze without charring and there will be no problm in your test result. But make sure that you sample becomes carbon free completely and no black particles is seen in the ash.

    @@poogylama1737 After charring, place the crucible inside the furnace immediately. Lid of crucible should be used to avoid loss of the ash content while burning inside the furnace. Desiccator is used when ashing is done until taking the final weight.

    This method is applicable for liquid sample but one more step should be followed. First of all, take your milk sample in the crucible and place on hot plate. Then heat the sample to dry it. After drying, follow the burning steps in furnace as described in this video.

    Why didn't you burn crucible in preparation of crucible .doesn't it change in weight of crucible if it burns before adding sample?

    You are right. Crucible should be burnt and cooled before taking sample in it. But we just dried in oven. Both can be done but burning is more appropriate.

    It is not necessary to make a sample free from moisture before ash analysis. Moisture indicates a quality of sample. So analyze your raw sample for ash content determination.

    Ashing is done for 2 hour at 700 degree Celsius. But if you burn the sample at lower than 700 degree Celsius then you need 4 hours of burning.

    This is only Ash determination. So we dont need to use H2SO4 in this test. If you looking for Acid Insoluble Ash test, please find from our video list.

    This is applicable for honey but the ash should not be in black colour. Burn again after adding 5 ml ethanol

    Thank you dear for asking... Ans: Total ash means total minerals. Ash content of any sample indicates the heavy metals and minerals content of that sample. During incineration in Ashing process, all of the compounds & elements are removed and become weigh-less except heavy metals and minerals. Amount of heavy metals and minerals indicates the quality of any sample.

    Richard X, high ash content food is good. High ash means high minerals. Minerals balance our body by different reaction and by maintaining ion channels.

    No. After taking Milk sample in crucible, it should be heated to dried up first. The. Transfer into furnace and do the same as in video
