Replica Plating - CSIR NET Life sciences | GATE Exam
Replica plating is a technique in which one or more secondary Petri plates containing different solid selective growth media are inoculated with the same colonies of microorganisms from a primary plate, reproducing the original spatial pattern of colonies. This is a screening method to isolated transformed cells from non transformed cells.

perfect but got one confusion. How do we get the ampR colonies back that contain our gene of interest after plating them on amp medium. I suppose they dies off

No they don't die we have to preserve them because we already know that they contain both self ligate and rDNA and the amp plate is called master plate and this process is also known as negative selection because genes which are not of our use are selected and then compared with the master plate to get the host containing rDNA

see when we are transferring the cells from master plate to the velvet, only some of the cells are getting transfered from each spot. So the master cell still have many cells on each spot. (since the cell size is small each spot contains lot of cells, which is not possible to show pictorially so you may get confuse).

In master plate, there are few bacteria reamains.Because we slightly press block on master plate. Then we compare both plates and find which colony took rcDNA.Finally collect those few bacteria remains in master plate.

but we have already plated simulatneoulsy plated them on thw woden block so by doing that the colnies will be in parallel and after the tetracycline is added to peterdush and all recominants die we can compare the petre dish and wodden block and pick out the colonies hope it helped (i also am a student so there might be a misktake)
