Neuroscience of Depression

About 1 out of every 14 people suffers from depression. What causes depression in the brain? Is depression inherited? And importantly, does depression medication work? In this video, we discuss the cause of depression, the changes in the brain, common disorders like major depressive disorder and persistent depressive disorder, and medicines used to treat depression.
When you are in the midst of it, you may think that depression is your fault, or that it is impossible to overcome depression. But that sadness and stress don’t have to rule your life. Here are some resources that may be able to help you or a loved one with depression:

    I've had depression since I was 17 but I had my first major episode while I was in the military. It was the scariest thing I've ever been through. I just don't want this to ruin my future. I have issues with zoning out now. Most of the depression is slowing fading but my cognitive abilities are terrible.

I will tell you I was breathing bc I had air in my lungs but I felt dead and drowning in despair. . He picked up my brokeness and making it new!     Depression is so debilitating. It makes you apathetic and tired all the time causing a lot of responsibilities and even basic hygiene to become neglected. This of course just makes depression worse and is really easy to get caught in a cycle. It's the worse.

    This is a great video, I had no idea about all the different possible causes of depression. While I have not experienced depression I have experienced anxiety attacks (most likely brought on by my thyroid disease). Personally I think the scary part is not knowing what is causing these unexplained emotions. When sadness or anxiety are caused by traumatic events - it's definitely not an easy thing to deal with, but at least you are aware of the cause. But when you are experiencing emotions without apparent cause, or disproportionate to the cause it can be really hard to deal with and makes you feel even more out of control. So when people tell you to just "snap out of it" it is even more frustrating. Thanks for a super informative video!

    Totally! I (Alie) have dealt with anxiety (we even made a video about it!) and it is really hard when you can't pinpoint the cause. I didn't even realize how bad I felt until I started getting care and didn't feel so bad anymore, because it seemed "normal" to feel that stressed all the time! I'm really grateful for the resources available to me, and to have really great professionals helping me navigate this time in my life; I don't know if I would have been able to handle everything life threw at me last year if it weren't for modern medicine.     antidepressants on their own have never worked on me, especially SSRIs. It took the combination of a stimulant (I have ADD) and an SARI I take for sleep to really kick depressions ass for me, and I had suffered for almost a decade and a half on incorrect meds. The addition of the stimulant changed my entire life. It even improved my sleep.

    I had some major depression a few years ago after ending a relationship. Getting dumped was fairly normal for me and I had strategies for dealing with it, but knowing what I put my ex-partner through by splitting with them pushed me over the edge. I was distracted and irritable for almost half a year until I went to my old pen&Paper group and took some time off work

    Great video! Definitely a solid introduction to the literature and handled with a lot of empathy. As someone who both researches and lives with depression neurobiology, it makes me glad that you guys are doing good work spreading knowledge! Do you think you'll ever cover the excitatory synapse hypothesis of depression? That's definitely my favorite explanation of MDD pathophysiology.

    Chronic stress causing depression makes perfect sense, unless you don't think its stressful to grow up in an unloving abusive household, rejected or bullied by friends, then oppressed by an unfair economic system. Shhhh theres a lot of people who don't want to talk about that stuff...

    depression is caused by emotional wounds as the person is emotionally hurt but in a hypnotic state of believeing he is just depressed and that there is no cause of the depression. i have experienced deep depression myself and know how that feels and know, someone injured my feelings in more than one way.

BALANCE and completeness go hand in hand, as imbalance and loss go together. The INTEGRATED EXTENSIVESS of thought (and description) is improved in the truly superior mind. Thoughtful, emotional, AND physical health, growth, development, and maturity matter. Indeed, thought, emotion, AND feeling are interactive. Warm yourself from within.     Yeah, the stigma is really strong in a lot of countries, including the US. It's hard for people to understand and empathize with depression. But we don't necessarily have the same difficulty with physical illnesses, for example. We just have to keep educating and breaking down that stigma towards mental health! Thanks a lot!

    Significant stressors lead to higher cortisol which uses up large quantities of b vitamins. B vitamins also help serotonin to re-establish the parasympathic response. Chronic stress, therefore, creates a feedback loop. I am in favour of the inflammation theory. SNRIs and SSRIs have also shown that they lower inflammation which explains why they take time to work despite the immediate increase of serotonin in the brain.

    Because of depression I lost everything I worked hard for. I worked hard and arrived in the USA. Then my depression was so severe I had to return to my home country to my family.

    Was diagnosed with dysthymic depression from early childhood some 15 yrs ago. Last year my diagnose was changed to persistent depression, as I don't have 'better' (less bad) periods anymore. I also have a neurological disfunction that's dopamine dependent, therefor I can't use SSRI's or TCA's, cause serotonin surpresses dopamin and I already don't have enough dopamin, that's why I use dopamine agonists. Been in therapy for many years, of which one in crisis care and one in cognitive group therapy. Made it worse rather than better. Used to smoke cannabis, which helped with the depression and the neurological disorder. Depression because it caused shorter short term memory, so I 'forgot' a lot of things to worry about and the other it helped with the spasms I have. Had to stop the cannabis, cause my car insurance threatened to stop my insurance if I continued using it. I haven't had a good night's rest for over 10 years. I'm lucky if I sleep 5 hrs, but mostly it's 2-4 hrs. Sleeping pills don't work or make me feel drowsy all day and I need to be able to drive. People depend on it. Don't have any hobbies anymore. Rode a bike for a long time, but had an accident in 2019, the physical damage forced me to stop riding. Just staying alive for others who depend on me.

    How does one distinguish between major depression and everyday, ordinary misery?

    "Ordinary misery" is a part of life that we all go through during our lifetimes. MDD (Major Depressive Disorder) is an ongoing organic pathology. It should never have been parceled out from physical health in the first place. It's not about grief, loss, etc, but when all the misery stacked up makes chemical changes in our brains. That can be treated; misery & grieving should decrease over time. Also, neurochemically, the neurotransmitters in the case of depression are dopamine, norepinephrine, & serotonin. There seems to be an inhibitor that prevents these neurotransmitters that cause the issue. Hope that helps. Take care.

    @@catmom1322 What if u have days where u can’t get out of bed and u just feel stuck, ur not really happy or sad ur just kinda numb, ur appetite changes from eating and never getting full to not eating because ur not hungry at all, some days you’ll be perfectly fine and then out of no where u just feel hopeless and kinda worthless? Is that just like normal misery or something else?

    I came here to try & understand what's happening to me, to my mind...& this has given me some kind of clarity, though the only time I experience mind chatter, is when it's extreme, it's almost constant, during/after a state of quite deep depression. The first time it happened many years ago, it frightened me as I couldn't switch off from it, it was a constant dialog of chatter, like commentary. Through my depression, I realised that I had nothing else to think about, as i'd lost interest in almost everything & life felt very uneventful, very boring. I'd lost interest in all the things I previously found interesting, exciting & worthy. After alot of stresses in life this past 2 years, i've unfortunately gone back into a depression & the same feelings, plus recently this mind chatter, which at times i've found very dibilatating. Luckily, as much as I don't like it, theres only been a few times were it's overwhelmed me, which has made me panic. In my first episode of it many years ago, it was so constant that I had to play loud music much of the time, to drown it out. I'm hoping that, as I come out of my depression & I gain back interests in things, in life...it will again subside. My understanding is that it comes on with me, when my mind is empty of thought, of what is usually my interests, yes even washing up & daily tasks. I call it an empty head & mind, that requires something to do...so it chats, mind chatter. I hope that this is understood to the professionals here & maybe helps others.     SSRIs can also have a negative impact on some people who experience depression who are also bipolar. In my case I was on the wrong meds for a year before being properly diagnosed. I've been thirsting for more science-based explanations of how complex depression can be, as I have learned somewhat how to control mania to a certain extent. When depression hits, though, it's like a ton of bricks.

    Its very strange. The neuroscience for depression and bipolar seem to be mostly unrelated to one another which is a bit counterintuitive. Thats why ssris have a very low success rate for those with bipolar and more new types of treatments like lithium seem to be exclusive to treating bipolar. No meds directly target the mechanisms for bipolar or depression, so they arent always really effective and can sometimes even work against us. Pretty oof

    This was interesting... I'd like to see an episode that focuses on bipolar disorder though... it's definitely a type of depression but it's certainly not at all the same as other types of depression. Side note. the background music was an interesting choice. Music that feels melancholy in a video about the neuroscience of depression... coincidence? me thinks not.

If a depressed patient have all established affects like very low serotonin & GABA, irreversible anatomical changes in brain etc. If that patient is at the brim of committing suicide.     I'm turning 19 this year, i've had depression sense I was 6 and self hearm sense 10, i've had medecin but nothing helped. So every day is hard and i go through it by thinking "one more day, just one more".

    "SSRIs work" ehh... I mean they tend to do more than nothing, but not enough that I wouldn't consider that statement false. They commonly have debilitating side effects like preventing virtually any sleep from occurring(and for anyone with a sex life, you can kiss that shit goodbye with a 50/50 chance), and shopping around to find one that isn't going to debilitate you just as much if not more than your depression can truly be hell if you're not lucky. They have legitimate uses, but we definitely overprescribe the hell out of them, and I'd recommend anyone to try non-drug therapy before getting pilled up. Also MAOIs are just horrible. They've been rarely prescribed for several decades for a reason, that reason being they violently interact with damn near anything, including several types of food. like there's a reason why virtually any ad for a drug will have "do not take X if you're currently taking MAOIs" in the fast speaking legal liability section.

I hope you’re doing well, thank you for the laugh about the kiss the sex life goodbye Seriously, that helped because it’s so true Also, that’s rather informative in your second section. Thank you for letting me know about MAOI’s. I will have to discuss that in the future. Also a fantastic tip regarding non drug induced therapy’s first. I noticed when I took medication I felt like I was changing as a whole, rather than changing the feeling of depression. I ended up feeling more angry and frustrated. Maybe I didn’t take the medication as I should or i wasn’t consistent. All I know is I do fine for a while when I have my “episodes” of what I want to call manic depression. But I wouldn’t know the difference with my lack of knowledge. I feel as though I am falling down a pit with no bottom, I see the light getting more and more distant. I have bounced back before, and it does feel better. I’m just not feeling it yet. I hope.

    I know I'm late to the conversation and I hope you're both doing ok The first paragraph is so true it triggered my nervous laughter, I had a full breakdown while on an SSRI and while for a short time my mood stabilised and I was more chill, I ended up getting so chilled I was numb to everything and ended up on a self destructive bender that nearly resulted in a suicide attempt. I took myself off my meds about 4 months after I got over the worst of it (I was forgetting to take them consistently towards the end anyway) and while I still have bad spells, I'm certainly glad to feel alive once again, even if it means I have to ride out the shitty times. Personally they didn't work for me but they can be a short term lifeline for those who need them. As I said, initially they worked wonders for me but after several months they wreaked havoc

    I was diagnosed yesterday with Anxiety Disorder after a couple of days of chest pain, difficulty in breathing and throat pain and insomnia. The doctor prescribed me 5mg of clonazepam sleeping pills for 4 days and a whole 10 mg of it in the 5th day just for me to sleep. That's for 5 days. And oscitalopram oxalate 1/2 tablet that's 5mg for 4 days then a whole of it 10 mg after for 2 weeks. My concern is i have looked all those meds up in the internet and I kinda the worried about the side effects that may occur especially that withdrawal syndrome side effect. I have also read some where that oscitalopram oxalate should be taken months or years thats almost part of a person's life and the side effects for me is pretty scary. Does this drug really is safe? I mean I can probably control my self without anti depressant drugs but the doctor requires me so. I dont want to be tied up with any of this drugs for a long period of time or even for a lifetime. Please if you have any advises that would be much appreciated.

    Hi Elvin! I (Alie) also have anxiety disorder, and I also take medication for it. I've even taken escitalopram in the past (though not currently). My personal experience with escitalopram didn't involve any major side effects or withdrawal issues, and usually doctors will work with you to scale up and then scale back down the dosage in order to make the transition as smooth as possible. I know that it's not uncommon to feel a bit "twitchy" when coming off of SSRIs, but the experiences I've heard about personally have indicated that it's not too bad and is usually gone in a few weeks. But it sounds like your doctor is prescribing an SSRI only for two weeks, which is very strange - most doctors would say that an SSRI takes 6-8 weeks to have any significant effect, and usually people will take them for at least several months. I have been taking a different but related medication (an SNRI) for almost two years and have overall found it to be very beneficial, though I don't plan on taking it forever. The doctors I have worked with have also been very considerate and we've talked a lot about the effects/side effects and adjusted dosage and medications as needed based on how I'm feeling - most doctors want to find the best solution for their patients, and not just prescribe a drug for the sake of writing out a prescription. And finally, I totally get where you're coming from with your concerns about the medications - a lot of the stuff you see online is very intense and often reflects very negative experiences. I think most people who have a positive experience just don't end up talking about it much. If you're not comfortable taking the medication you've been prescribed, you might want to try to see another doctor and ask for another opinion. I'm sorry to hear that you've been suffering with such intense symptoms from your anxiety lately; you may find that the short term medications and sleeping pills are enough. If you're not interested in medications, you should also ask about therapy or counseling - it takes longer to see a change, in my opinion, but therapy is very effective for managing anxiety for many people. I hope this is somewhat helpful!

    Thank you for the quick reply. I used to be a happy man though since when I was a kid I have this panic like feels everytime I face unknown people when I am reporting or performing to a class(or a lot of unknown people). Not until yesterday that I knew that i have this kind of illness which I don't really expect to gain. My doctor said some cases (not mine) could last up to 6 months though. We will meet again after two weeks to assess my progress. That must be the reason why he only put 2 weeks on my prescription. I hope you understand my poor english. I really appreciate your reply and the knowledge that you share with me.

    Clonazepam can cause a severe withdrawal syndrome if discontinued abruptly. Never decrease your dose or stop without consulting your doctor first (especially if you intend on using clonazepam longer-term, like >2 weeks). Escitalopram, on the other hand, may cause a mild-to-moderate withdrawal in some people but it's relatively uncommon and mild compared to most drugs. Both are generally safe drugs otherwise, side effects if they occur are usually mild. If you're reading the side effects off the medication guide I'd recommend avoiding that... it can definitely cause a lot of anxiety, I know from experience. Just remember that serious side effects are extremely rare (under 0.1% if I had to guesstimate) and that these medications are extensively tested first to make sure that they are safe.

    Just stumbled across your channel, and I LOVE what you're doing. I have a mental health channel as well, and I talk about neuroscience quite a bit. Have any of you read the new book Lost Connections by Johann Hari about depression? It's really fascinating. There's a lot of talk about anti-depressant medications and how the success rates aren't nearly as high as people think. It goes over a lot of the other environmental and societal causes of depression as well as solutions. Anyways, I just subscribed and would love to collab with you some time if you're interested! I'm a recovering drug addict and do a lot of research on the neuroscience of addiction, so I'd be down to do a guest video on that if you'd like!

    Great question. Diagnoses change over time. The way I see it, it's sort of like a physical health diagnosis. People recover from their illnesses if they receive proper treatment. However, you may be more susceptible to it in the future if you've had it in the past. It just depends! Regardless, if you're struggling right now, get some outside help. You don't have to be alone in this!

    radve. I felt like this when I was 12, I’m now 16 and it’s just gradually got worse. You should try and seek help before it gets too bad as being a teenager does make it worse.

    What you are feeling is valid and may get more severe if you don't do something about it now. Mention the way you're feeling to a trusted adult and if you work with it now, your symptoms could be nothing more than a bad time and not a lifetime illness. My mistake was not getting help when it first started, and it only gets worse. If only I knew then...

    Thumbnail: Doctor, I don't feel so good. Imagine depression as a car with a failing engine! It drives suboptimal, but ok... Makes weird noises or fumes from time to time... You don't realize how much some parts struggle until they suddenly break. Depression comes from misconceptions and similar effects. Everything is fine! They say... When I realized my depression at 11yrs, of course people didn't believe me - and I believed them. So I searched for my problems elsewhere... Now I'll get 30yrs. And I realized how often I was right... This unintended gaslighting almost destroyed my life 😬

    sadness is normally a sign the depression is lifting, there isn't much feeling during... i'll watch something and laugh cry, and that's start of recovery... i hate the word depression, it was a bad choice of a label... i think there are times medication will work for some people, but i think focus should be on identifying the problem and treating symptoms as a last resort... 3 weeks for an anti to take effect, means it is, just as it isn't needed any more... better a tranq... with shell shock being fairly indiscriminatory, and coming out of my first major ontop of dystemia... i have become very motivated, and neuroscience ties a lot of it together and i can get my own data now... and 20 years of other data open... the last effort in trying to understand my psychology, then compare that to the pattern of my behaviours, helps me to avoid it in the future... other than bipolor, i think a lot of depression is caused by the intersection of stimulus against psychology over time... sometimes it builds to a break like dystemia, or hits like a sledgehammer like the major did... all that mental pain, is a mental threat warning system like physical pain... it is much harder to see the threat, which is why behaviour pattern recognition helps, and its not the same for everyone.... SCARF has helped me to frame the work and some other components... also being an empath makes me more likely to be a canary in the coal mine, and can become a target of narcissists sometimes... also being a 'night watchmen in the caves' means i allocate energy to fit into other peoples schedules etc... and a few other oddities on their own aren't an issue but combined with stiigma has made things a lot harder than required... but no shame, it can happen to anyone...

    i would say people in the same family, share similar psychology, and will probably also receive similar stimulus, sometimes in negative feedback loops with each other... that imbalance will cause depression eventually... i don't think the depression itself is genetic necessarily...

    Hey Evert! ECT is only used in very extreme cases of depression. We are still not sure exactly how it works or why it works. Back in the day, it used to be a front-line treatment for major depression. It did a lot of harm to people as a result. I think you would have a hard time finding anyone who would prescribe it now. Doctors and mental health professionals today will pursue medication or therapy as treatment instead.

    I take snri at a high dose and it helps a lot. I also take a controversial natural substance and it truly is saving my life. I only started taking it as a last resort. It actually made me go from atheist to agnostic. I think indigenous people are laughing at us when it comes to mental illness treatment.

    Routinely do not sleep well at night and wake up feeling very depressed. I ate two hamburgers and a banana right before bed last night. Slept better and woke up feeling amazingly better. (More energy, eager to get up and virtually no depressed feeling.) Any clue as to why?

    Abe Matt You would know if you suffer from DEPRESSION. Many people, even when receiving 'therapy' are misdiagnosed as being sad or temporarily affected by inability to cope due to a particular instance. This may be due to the person suffering from deoression being unreceptive to questions being fired at them, etc., and just wants to go home and pull a blanket over their head. Until a little respite may be available.

My conversations with people who have depression have indicated that sometimes it can make you feel like you're not behaving the "right" way and that you should just "shake it off" and get back to normal. This can add to the sensation of feeling worthless, because even though you know that you /should/ just get up and act normal, you can't get yourself going. If you aren't sure whether or not you have depression, you might want to think about seeing a doctor or a psychologist to talk about your situation and symptoms - they can help you determine the best course of action. And if you have already been diagnosed with depression but aren't sure you really have it, it might be worthwhile to get a second opinion from a doctor or therapist or consider alternate treatment options. Everyone deserves to enjoy life, so we hope that you are able to navigate this situation and feel better soon!

    Abe Matt I've been asking myself that. I grew up in a "no fear", "always work hard for your money", "don't mess with me and I won't mess with you" kind of environment... However, there is always some jerk off out there who gets his rocks off by messing with someone who is in a more vulnerable position than said someone is... They prey on these people until they suck the life out of them... But you know what?? Screw that crap! We do NOT deserve that kind of treatment from anyone... Bro... Or madam, be stronger than you ever have been, nerds, people who aren't as physically strong as the rest of the cheerleaders and jocks at school, people who feel the need to leave school early because some gang member is going to hurt them.... All awesome people with a level head, let's just take over!

    Nearly 40 Trillion Human cells make up the body but hardly make up the majority of mass. The number of cellular Microbial non-human cells that are facilitated by the body are estimated to outnumber human cells 10:1. Why is this important?
