    The explanation itself is great! However, the bullet points at around 2:40 are very confusing. Why are ammonium sulfate and sodium sulfate listed as two separate points, they should be examples of point 2. Point 1 should be a catagory under which points 2-4 fall. Point 5 should also be its own catagory and of course 7 is an example of 6 and 8 (higher temperatures) should be a point in the catagory of 5. Everything else however, is very concise and you explain it very well, so thank you!

    this explanation video is the best one for HIC! I saw many, and only this one explained everything in detail I.e step by step not just the gist of the technique. I know HIC is based on hydrophobic interactions but I need to know the steps and this one explained everything very well 👍

    please if you can explain to me, why the mobile phases containing a concentration of ammonium sulfate promote the binding of many proteins to HIC columns??

    Kosmotrophs are such molecules that interacts with water molecules. When ammonium sulfate solution is introduced, the water molecules that is bound to ligands, get detached. And ammonium salfate-water complex washed away from the column. So, the proteins can bind with the ligand according to its hydrophobicity since the proteins now have access to the naked ligand.

    Chemistry explanation is great, but can I get an answer to Why? Why is this assay performed? What are we trying to find out about the molecules? And for the love of God please don't tell me "the hydrophobicity of the protein", like why is that important?

    Speed of word is perfect, still if you have problem then watch the video at lower speed by changing the settings of the video (three dots on the upper right corner).
