    You need to be careful when heating cheap hardware like those nuts and washers. They are zinc coated, and the games that heat releases from zinc coatings are toxic. You can do it, but only safely with a respirator or at least outside. A safer method would be JB weld or some other epoxy or glue.

    The way I cut my sections is as below: Snap a shaving blade and sandwich it with two new non-snaped blades, then put the whole contraption in the spring loaded binder paper clip and press the blades in to the specimen. Afterwards release the clip and you have a very thin section between the two outer blades, as thin as the middle blade thickness. It works quite well as long as the blades are sharp. Also make sure the blades don't bend while you pressing then in to the specimen.

    if you added a spring between the wing nut and the nutted washer, it would preload the screw, minimizing the backlash or wobble.

    You should have probably sanded down the washer before trying to solder it, the galvanization on the washer would have prevented it from attaching properly. Then maybe on the other side the part you're actually cutting on, consider trying to sand it down smooth using a wet sander and get it to as close to a polish finish as you can. It's not hard or expensive to do. That should improve the quality of your cuts by a lot. A double-edge razor blade broken in half can be used as a super-thin knife, when broken into two they become sturdy enough to put a bit more force into them without bending. Another thing you could do is use graphite from a pencil to draw a tiny bit onto the surface, Graphite works as a dry lubricant that will make it more easy to slide the blade along. If your washer is rough it won't catch the blade as much. Although that can contaminate the specimen with graphite shavings. I mean, this is a DIY project nothings perfect.

    If you sand it, the surface might damage the slice, like if your skin were dragged across concrete. The slice he made was quite fine as far as the carrot was concerned. Most samples (like biopsied tissues) are embedded in wax (paraffin, I think?) so that the target tissue doesn't bend like it did in the carrot. It is easier to hold a paraffin slice and thus get a thin slice of the pine needle. This is an excellent, cheap, usable, and minimal design. I give it five stars out of five! I might be using this if I ever get a nice microscope! Now I need to find a good embedding method.

    Cool. Another thought: After watching the video, I looked at a few hand microtomes on the net. As a hobbyist, I immediately thought of a micrometer screw. They cost between 10 and 25 euros. Actually, one should be able to convert even the cheapest ones relatively easily, still remain far below the new price of a hand microtome and on top of that, get a device with which one can make precisely measured cuts, like with a real instrument.

    Using a hardened, metric fine thread, machine bolt and nut may also have better tolerances, readily available at hardware stores, and they actually have "cut" threads, as opposed type of screw that you used...which often have "rolled threads", with wide tolerances (loose fit). There are also "stop nuts" that have a little piece of nylon in one end meant to keep nuts from working loose, those would take up "backlash" that makes screws wobble in nuts. Stop nuts are a little hard to turn at first, but if you run the nut all the the way up the bolt and then back them off, they turn easier and "seat" on the bolt. You can also find them in a wide range of diameters, which means you could get one that fits snugly within the bore of your DIY micro-tome. A variety of "knobs" are available that could make a DIY with stop nut easy to turn, stays in place, and could be marked with "guestimates" for thin vs thick slices. Two part epoxy is a great way to fix items together and make smooth finish.     WONDERFUL!! I love the carrot trick. I want to make a bunch of cuts of an onion root tip to see mitosis and this just might be the ticket for that!

    As I recall, in my Biology 102 lab we had just a simple diy microtome consisting of a small section of metal tube, some soft foam sticks the same diameter as the tube but longer, and a utility knife. Advancing the specimen through the tube was done manually by pushing the foam up from the bottom and took a fair bit of practice to get the appropriately thin sections and yes, the foam filled the blades quite quickly. Still, the cost was negligible. This technique would appear to be an improvement on my simple tube. 👍

    Don't use a stiff bladed knife. A single edge razor blade is best. The blade is flexible so it gives a sharp uniform cut.

    Perhaps use epoxy like E-6000 instead of hot melt? It's easier to remove the excess (room temperature). The small washer with points on the bottom is a flange, I think. Use an X-acto knife for your blade. It is meant to be used as a very sharp blade. You can get capsules with about 11 blades in each. And then get a handle to hold the blade and you are ready for excellent results, or -as my family says -you're cooking with gas!

    auch hier wird der Fehler gemacht das Messer auf einer rauen Metallflaeche zu fuehren, was dazu fuehrt dass das Messer sofort unscharf wird

    kaufen sie sich doch mal ein gebruchtes Ultramikrotom und machen Glasmesser. Die kosten etwa 300 Euro und vergessen sie diese Spielzeuge. microtome toys
