How does DNA look like ? Agarose gel electrophoresis

Agarose gel electrophoresis is used to resolve DNA fragments on the basis of their molecular weight. Smaller fragments migrate faster than larger ones; the distance migrated on the gel varies inversely with the logarithm of the molecular weight. The size of fragments can therefore be determined by calibrating the gel, using known size standards, and comparing the distance the unknown fragment has migrated. This technique can be used to resolve complex DNAs (i.e., genomic DNA)

    Does smears in the result show that we have a lot of contamination in our sample? We take 2 microliters of loading dye and 3 microliters of DNA .. so we have to change the setting of pipette to 5 microliters everytime we load them?

    You can go for Rnase treatment or do some changes in protocol. Concentration of dye and DNA does not affect the quality, smears shows that the problem lies in protocol itself. Either protein or RNA must not have been removed completely.

    The size of the DNA molecules can be estimated by comparing the position of the bands on the gel to a standard ladder of known sizes. The smaller DNA fragments will travel further down the gel, while larger fragments will move more slowly and remain closer to the top. The intensity of the bands can provide information about the amount of DNA present in the sample. More intense bands indicate a higher concentration of DNA.
