Ans 3 mam ....first use leu- his+ then use leu-his- in replica plating method

Ma'am answer is C, but 1st plate we will get the self ligated ones, and in replica plate the recombinant ones.

While performing replica plating how can we use dead bacterial cells ( recombinant transformed ) for further experiments ? If these cells don't have tetracycline resistant gene then they will die ..

Good question ! We usually have large stock .so. After knowing through replica - plating we mark the stock as competent cell stock .they can be used for further experiments.
