Its a long task. Its difficult to explain here. Neverthless you can subculture your colonies on different selective & differential culture media like MSA, Cetrimide Agar, XLD, SS agar, Chromogenic Coliform Agar, MRS, MYP agar, etc. After that do biochemical tests of the selected colonies for identification.

Hi! I just wanna ask if it is necessary to use both RVS and MKTTn for selective enrichment? as well as XLD and BSA for Isolation? thank you so much

Yes. Both needed for selective broth. And you can use only XLD for isolation

No. Peptone water is used for Non-selective enrichment. Non-selective enrichment can not be achieved by distilled water. So you must use Peptone Water.

What are the advantages and disadvantages of this method as compared to others detection methods?

1. Advantage: This method involves double enrichment steps which increases detection possibility and facilitates to detect Salmonella more selectively, thus this is the more accurate method compared with other methods. 2. Disadvantage: MKTTN media, frequently, becomes not available in local market and there is no replacement media of it.

Don't cover the lid of plate until medium is solidified N make sure the agar is completely dissolved in Duran bottle while heating in microwave This can be achieved when u see the medium is boiling clearly i.e bubbles formation.
Wauu. This is really a good video. Can anyone share with me a full document where I can find medium types used to enumerate bacteria founds in food. Please help if possible. Have a nice weekend

Why does solid foods need 24hrs for incubation after homogenized with buffered peptone water???..Not only for salmonella detection but also for enumeration of TPC??

24 hours incubation in buffered peptone water is needed for the enrichment of salmonella. But this enrichment is not applicable for TABC or TPC

Do we need to make 225ml of BPW in the non-selective isolation stage? Why not less than that?
Pls why did u use 1ml of peptone in mkTTn broth and 0.1ml in RVS broth
