  I have a question, the powder that i have used for the maceration can i still reuse it? After filtering my solution, there was still leftover solute. After some time, it became dry again. And so i was hoping if i could use it again to make another batch of the maceration process. My plant material is very limited, you see. I used methanol as my solvent btw

  You can but the amount of bioactive compounds might be lesser than the first batch.

I have another question : for ex if we use ethanol-water as solvent, after the extraction we eleminate the ethanol first using ratary-evaporator,but how we eliminate water fraction ?   Hello! Thank you for this informative video. I have 2 questions to which I hope to have an answer to, it would be very much appreciated. 1- In our lab, we don't have a freeze dryer (lyophilisateur), and in a lot of references, i read that they used a drying oven at 40°C for 48h to evaporate to water in the extract. It is okay? Will it not affect my results too much? 2- For your alcohol extract, after using the rotavapor, the extract was liquid meaning there was still a little bit of solvent in it no? if i'm right, how will you then calculate the concentration if you don't know the initial concentration of your extract? Thank you!

  1. Yes, you can do that. 2. Yea, there will be some alcohol left. But you need to further evaporate this in a fumehood for a couple of days. Concentration of what? For what? It is automatic that the concentration of the extract you have collected is 100% pure crude extract. If you want to use it as a treatment for testing, then compute the specific concentration you need for the testing using weight of the extract/volume of solvent.

  Hello po! Thank you po for the informative video! It really helped us understand the methodology needed to conduct our research. May we ask po how much plant material (in grams) do we need to use to extract 5mg, 16.25mg, 35mg of the pure extract? And also how much solvent is required for maceration? We are currently struggling to compute the amount of grams of plant material to get the according milligrams (We tried using the vague references in the video but still struggled). Also po we need to compute the amount of solvent used for maceration and the ratio is 1:2 (w/v). The volume of our final solution is 50ml and we need to dilute it for 3 concentrations (10%, 32.5%, 70%). After solving this, the grams needed for the dilution is already stated above (5mg, 16.25mg, 35mg). To conclude, our group wants to know how much grams of plant material is needed for the final product using the 1:2 ratio for maceration and how much solvent is required. **We know that this request is a bit hard, if it cannot be answered, do you have any suggestions po on how we can compute it?** Thanks po doc!!

  @@LVCAS_MAGNVS then use retinoic acid or quercetin if you want. They may have antiangiogenic properties but these are not drugs used for angiogenesis or cancer.

  Contaminated incubator, not properly cleaned, not properly closed, not airtight door, etc. Better clean your incubator first before using. Maintain 37C temperature and 55% relative humidity.

  In this experiment (nasa video), I used various extraction solvent. Crude extract ang tawag sa makukuha mong extract after the extraction. Kung ang gamit mo is ethanol, you’ll call it ethanolic crude extract. If water, you’ll call it aqueous crude extract. Maceration takes 3-7 days, depending on your research design. The actual extraction for ethanolic extract takes less than an hour. For aqueous extract, you only need to boil them. Filter. Collect the supernatant and freeze it. Once frozen, load it in a freeze dryer. It will take 1-2 days or less depending on your extract.

  Hello po. We are currently conducting a CAM Assay about a plant's anti-angiogenic effect and all of our eggs died po. How can we make them survive for at least a day para lang maobserve namin yung blood vessels nila and macount po? And I have learned from your reply here sa comment section about the hemoglobin test level, paano po yon?

  You should monitor all conditions properly, the humidity, temperature, etc. Measuring the Hemoglobin level is an alternative way of quantifying the angiogenesis. If mataas ang hemoglobin level, pro-angiogenesis. If mababa, anti-angiogenesis. But this is strictly for CAM assay with tumor implants. If walang tumor implant, you can use the commonly used technique, which is counting the 1st, 2nd, and 3rd branching points of the blood vessels.

  You can dissolve those in the flask with your extraction solvent. Put them in an evaporating dish and place them in the fume hood for a couple of days.

  Rpm doesn’t really matter. This depends on how fast you want to remove your solvent. Temperature depends on the rotavap’s pressure, and the boiling point of your target bioactive compounds (not the plant extract).

  Hi doc! Our study po is the effectiveness of c. ternatea linn in lowering blood sugar of alloxan-induced rats. We’ve chosen po the decoction method to extract the C.ternatea. Can I ask po what specific temp and for how long we will boil the C.ternatea? And, what can we use po to filter the extract after boiling?

  For the temperature, it depends on what bioactive compounds with sugar-lowering properties you want to extract from the plant. But in general as long as it doesn’t rich 100C to prevent the bioactive compounds from degradation would be enough. You can use a filter paper afterwards to separate the plant material from the extract.
