Hey! Are you even curious about how MX3 gets "xanthone" from mangosteen or how Myra-E extracts "d-alpha tocopherol (vitamin E)" from soybean oil? Typically, we see in the advertisements of whitening, antiaging, and food supplement products the terms like "ascorbic acid (vitamin C)" in Ceelin, "resveratrol" in Slenda, or "glutathione" in Vitapack Megawhite. These terms are names of bioactive compounds that exhibit tons of health benefits like antioxidant, anti-inflammatory, antibacterial, antifungal, antidiabetic, antiviral, and even anti-cancer properties.

    2:43 please.. have a question.. what type of solvent you have used for maceration ? I have another question : for ex if we use ethanol-water as solvent, after the extraction we eleminate the ethanol first using ratary-evaporator,but how we eliminate water fraction ?     I'm a 4th year pharmacy student and want to be involved with pharmaceutical science research. I've been thinking about taking a Masters in Pharmacy, but I'm second guessing because from what I see the type of research produced is usually assays of plant extracts which I kinda don't like. The type of research I want to do is along the lines of polymers, nanotech, etc. (i haven't quite figured it out yet). Do you have any advice for me?

    By “fortify” do you mean purify? You need to do chromatic profiling, separation and purification techniques for that.

    Hello! Thank you for your comment. To answer your questions, 1st. the reason behind freeze drying is to remove the water content from your extract without deteriorating your bioactive compounds. 2nd. if you want to obtain a liquid form of your extract, it's either you can dissolve the powder in your treatment vehicle (solvent used treatment, depending on your experiment), or you can use the extract automatically after decoction and filtration, but you have to make it more concentrated. If you don't concentrate your extract, it will probably yield lesser bioactivity.

    Preferably dry the fruit to saturate the concentration of anthocyanin. Since anthocyanins are light sensitive make sure to dry it on a dark areas.

    There’s no need to follow any ratio. As long as you have totally submerged your plant sample in your solvent then you’re good to go. After all, will remove the solvent when you run your mixture in a rotary evaporator.

    @@kaeyaalberich4427 you just have to weigh certain amount of your extract and dissolve it in a certain volume of solvent medium. Example: 10mg of plant extract dissolve in 10mL water will make 1 mg/mL plant extract.

    What is the shelf life of the "extracts" when performing extraction process? In my case, I consider Iyphilization/freeze drying the plant to achieve the same texture of green tea before boiling it. I wanted to store a large amount of dried leaves for future purposes and save it for decoction. Will it affect the shelf life?

    Around 1-3 days.. it depends.. you’ll know it’s ready if you can’t smell the solvent from your extract and you can observe that it has dried up.

    Hi Doc! We are currently working on a research where are going to extract Peroxidase enzyme from papaya to detect glucose in a urine through GOD-POD method, but we are struggling in finding a laboratory that will help us to extract the Peroxidase from Papaya so that we can have a analytical grade peroxidase that we can use for our research.     Hello! Thank you for this informative video. I have 2 questions to which I hope to have an answer to, it would be very much appreciated. 1- In our lab, we don't have a freeze dryer (lyophilisateur), and in a lot of references, i read that they used a drying oven at 40°C for 48h to evaporate to water in the extract. It is okay? Will it not affect my results too much? 2- For your alcohol extract, after using the rotavapor, the extract was liquid meaning there was still a little bit of solvent in it no? if i'm right, how will you then calculate the concentration if you don't know the initial concentration of your extract? Thank you!

    1. Yes, you can do that. 2. Yea, there will be some alcohol left. But you need to further evaporate this in a fumehood for a couple of days. Concentration of what? For what? It is automatic that the concentration of the extract you have collected is 100% pure crude extract. If you want to use it as a treatment for testing, then compute the specific concentration you need for the testing using weight of the extract/volume of solvent.

    I’m not sure but I think you need to perform steam distillation to get extract d-limonene.. try checking then form of D-limonene and look what steam distillation is. 😊

    Water is more polar than ethanol. If you want to extract bioactive compounds specifically those with antimicrobial properties, better check the literature of what Antimicrobial compounds present in your plant and identify in which solvent are they soluble.

    Hai, when you freeze dry. Is it ok to fill almost full or leave a small space in your centrifuge tube? Because the technician in the lab that I'm using freeze dryer would tell to fill just half. but I filled the same as in your video. Thanks.

    There is no recommended ratio of solid to solvent when performing maceration because the end step is totally evaporating the solvent. So, as long as you have completely submerged your plant sample in your solvent then it’s good.

    1 and 2. You can only use water in decoction method. You’ll be extracting flavonoids and phenolic compounds that are soluble in water only.

    Hello Dr. JP hope all is well, I was wondering if there is a less destructive method of extraction there. All of this heating and drying surely would destroy much of the plant original compounds leaving us is a big chunk of oxidized and a lower grad less therapeutic extract...     Personally, I agree with you. However, I don’t thinking you’re making too much alterations or destructions to the bioactive compounds as long as you are in the limits. Hence, it is important to really know the your target bioactive compounds, such as their boiling point, melting point, polarity, solubility, etc.

    You can try decoction method. However, using this method you can only extract water soluble bioactive compounds.

    I have a stupid question but please help, i want to extract nutrients from Seaweed to use it as fertilizer does boiling water denature or destroy any useful amino acids or plant hormones like auxins or cytokinin etc?

    Before introducing you treatment (extract) to animal models you should make sure that all of the extraction solvent (ethanol) has completely evaporated. Usually, you can administer your treatment using the common drug delivery methods like oral, intravenous, intraperitoneal, etc. You can also add your treatment to the feed formulation but it will depend on what kind of feeds you have and its nature (dry/wet). Moreover, adding your treatment to the feed formulation might lead to inaccuracies in the volume or concentration of the drug (extract). So, you have to take note of that too. Like for example, if you want to treat your animal model with 200uL of 10 mg/kg extract, you plan how you can administer this accurately to all of your replicates to avoid biases and such.

    Use lesser heat as possible to prevent the compounds from degradation. For isolation, prepare your samples as clean and as accurate as possible.

    your rot' evap' setup ... a few questions : 1. what is that thing the sample flask attaches to ? it looks like some sort of splash - guard and does it have a part number ? 2. the condenser glass thing ... does it have a part number too ? you did well with the camera work to capture the p/n's for the rotavapor and heat - bath. with all these p/n's one will be able to shop for them

    Hi! If you have no plan on retrieving your extraction solvent (alcohol), you can just use a typical water bath. Maintain a certain temperature, use the boiling point of your solvent or somewhere close to that temperature. Then continuously stir your extract during the evaporation process.

    Maceration is simply soaking your plant material in your extraction solvent, like ethanol, methanol, etc for a couple of days. For the temperature and speed of the rotary evaporator, the temperature depends on the boiling point of your extraction solvent and the pressure of your vacuum (vacuum is part of the rotavap).

    Yes, you can. But first, identify your target anticancer compounds. Next, determine in which solvent they are soluble. Then use that solvent to extract your compounds.
