The Amazing World of Microbes

Ғылым және технология

Hello!
This is James, and as many of you already know, I am crazy about the microscopic world. I will turn 30 on March 19th and wanted to start a campaign to get my dream microscope. I know many of you people love my videos and want me to make even better ones! So you can support the campign to get some cute merchandise! Thank you!

Why does it always seem like these guys are 2 dimensional? Is it because they're under a slide and have limited movement because of that? It always seems like we have side profiles, but not all other orientations. I just wasn't sure if that was because of the way the Microbes are built or limitations of the Microscope slides. Great video either way!

Yeah, they're basically limited to 2D movement.

You can actually see rotation and difference in distance in these, you just gotta really pay attention, I think the see through body give the illusion that they're 2D

Hello my fellow Micronaut, I absolutely LOVE your content, THANK YOU SO MUCH for sharing your expeditions into the Microcosmos, you have me HOOKED! I've been lurking here and on JTMC for a few weeks now, while I was waiting on my first decent quality scope, which is one of the reasons I'm commenting. We got an AmScope T690C-PL Trinocular Compound Microscope, with 4 Infinity Plan Objectives (4X, 10X, 40X, & 100X Spring/Oil). Do you know anything about this scope? There is a guy on the Microbehunter.com forum who owns several quality scopes, with his most expensive being a $3000+ Olympus, and in comparison to the AmScope T690C-PL, he said that he can't tell the difference between the image quality of the optics, not only that, but the action of the stage is actually a bit smoother on the AmScope. I just started using the scope yesterday, and admittedly, it's been a bit of a learning curve. I've never used a scope of this caliber before, the Kohler Halogen Illumination System (along with the 3D-Stage, condenser/diaphragm & Oil Immersion) is all very new to me. I think I may have setup the Kohler Illumination incorrectly while trying to calibrate it, although I'm not sure. I can still see microbes at all magnifications, everything at 40X and below is crystal clear with these "Infinity Plan Objectives" (I didn't realize just how TINY some of them are, I had trouble seeing some of them even at 1000X, its CRAZY!), but the condenser needs to be pretty low below the slide to get the right focal plain, if I raise the condenser to within a few mm from the slide and try to focus on the slide/specimen, then it actually focuses on the frosted diffuser lense that sits directly above the halogen bulb, looks kinda like static on a TV screen with no signal. When I was "attempting" to setup/calibrate the Kohler Illumination I don't think I did it correctly, as I was using the field diaphragm instead of the condenser. With the field diaphragm closed to 1/3 field of view, I focused on both the edges of the field diaphragm and the frosted base/bottom lens simultaneously, why, I don't know, I just got my wires crossed on how to do this properly and I'm not sure if I did something wrong that I need to fix/correct/re-adjust. If you could PLEASE offer some advice on this I would be VERY GRATEFUL! We have a scope camera on the way, so I could actually make a video showing you exactly what I'm talking about, if that would help you diagnose my issue, if I even have an issue, I'm so confused on some of this stuff. As I said, I "CAN" see microbes at all magnification levels, I just have a feeling that I focused on the wrong focal plain during setup of the Kohler Illumination system. We bought this scope as an "Open Box Item" for $700 through AmScope on eBay, which apparently means that it was either purchased and returned, or was a display model, either way they claim that it has been thoroughly inspected, tested and is in 100% perfect working condition (they do seem to have good customer service, and offer a 5yr warranty, but I digress). The problem is that, I don't know what "Perfect working condition" even is on a scope like this, so I have nothing to compare it to. As of Wednesday-8th, I have 30 days to return it for a full refund, so I want to make absolutely SURE that this scope is in A-1 condition and performs to the manufacturer's specifications before then. The main purpose of this cope is for R&D on a new AACT (Actively Aerated Compost Tea) formula, and I have several sources of microorganisms (mainly aerobic/probiotic, but some anaerobic as well) that I would be more than happy to send you samples of. You might be surprised as to what biology my vermiculture contains (worm castings, I feed them a very high quality diet of wholesome, organic, non-gmo ingredients), as well as my static compost and fungal dominant wood chip compost. When it comes to growing plants organically (the way nature intended it to be), it's ALL ABOUT THE SOIL BIOLOGY! Here's the scope we purchased, please let me know what your thoughts are on this model? Should we keep it (all the important components are upgradable, should we want to get darkfield, phase contrast, etc) or return it and get a different model? Our budget is $800, which I know isn't much in the way of quality optics, but it's what we have to work with. So if you know of a better scope for the price that you would recommend over the AmScope, please let us know. Thanks for your time, stay healthy and we look forward to hearing back from you. www.amscope.com/40x-2500x-professional-infinity-plan-achromatic-trinocular-compound-microscope.html#product_tabs_review_tabbed

Hey William! Thank you so much! I can, of course, help you with that! The problem you are talking about is well-known with these types of budget microscopes. When the condenser is at some certain position you see the defects on the glass right above the light source and it's not connected with the Kohler setting, I found a couple of ways of solving this issue. The easiest one is by lowering the condenser a bit down instead of keeping it at the highest position. First, use the 10x objective and set the condenser to the highest position possible, keep the field-diaphragm full open and close the diaphragm on your condenser quite a bit so the light is not too bright but not too dim as well. Then slowly lower the position of the condenser until the spots disappear. If you don't see the spots and the image is still not clear enough the problem might have been caused by the sample preparation, when you prepare the slide, try to put just a tiny drop of water so the distance between the slide and the coverslip is tiny and you get a better plane to focus. And if you are using a 100x objective you need to use the immersion oil, otherwise, all the images will be hazy because these oil objectives are not designed to work without the oil. We made our first three episodes on JttM with the exact same model, AmScope B120 uses, just a non-branded version of it. It didn't even have infinity objectives, I bought this microscope for $160 almost 3 years ago. First of all, I should say that microscopy is an art and a microscope is just a tool. Yes, there are major differences between the cheap ones and the expensive ones from companies like Zeiss, Leica, Nikon and Olympus but overall anyone can get a decent image out of any microscope with practice, and I am a good example for that. Please, don't give up and you'll see what I mean with practice. Also, feel free to contact me via email and send me updates! jam.and.germs@gmail.com Best, James

Let's go on a quest of discovery, into the vast, infinite realm of the MicroCosmos. Through persistence, perseverance and sheer determination (not to mention some very, VERY EXPENSIVE equipment. Anyone in the position to do so, please help James get the tools/instruments he needs to do what he does.
