Romulus takes place after the first movie but before the second.

This is after Ripley destroyed the Nostromo in Alien (1979), but before she's found in hypersleep 50-something years later in Aliens (1986). It takes place during that 50-something time frame that occurs between Alien and Aliens. A lot of Alien media, like comics and novels and etc., take place during that time frame too. This is the first official canon movie to do so.
