If you watch a recent Kurzgesagt video on Exercise, you will find that apparently exercise reduces inflammation as it uses energy on something other than inflammation! This would then explain why Exercise helps with NAD levels are reducing inflammation would naturally increase NAD, but the Doctor did not mention and perhaps wasn't/isn't aware of what the video explains, worth checking out!

She may be on to something, but all the ingredients contained in her formulation are readily available at a fraction of the price. I'm not convinced that nicotinamide does much to increase NAD levels, at least not according to the research I've seen. Also, some actual research or studies involving her products would be appreciated.

I prefer nicotinic acids flushing niacin as it is very effective at increasing NAD and it reduces LDL and increases good HDL cholesterol. Whereas nmn and Nicotinamide has opposite effects on chol levels, per my understanding

🎯 Key points for quick navigation: 00:00:00 *🧬 Dr. Conlon's background and motivation for founding Neo Labs* - Dr. Conlon discusses her scientific background in bioavailability and drug development. - She explains her transition into longevity research due to the potential impact on age-related diseases. - Highlights frustrations with the slow pace of drug development and the underutilization of natural compounds in research. 00:06:26 *🌿 Understanding the role of NAD in aging and Neo's approach* - NAD's crucial role in cellular energy production and repair processes. - Discussion on the decline of NAD with age and its impact on cellular health. - Neo's strategy focuses on addressing the root causes of NAD decline through the Salvage pathway and optimizing nicotinamide recycling. 23:31 *🧬 Nicotinamide Metabolism and Cellular Function* - Nicotinamide is regulated in cells through methylation enzymes like NNMT, which attach methyl groups and signal its excretion to prevent cellular buildup. - Studies indicate that physiological levels of nicotinamide do not inhibit NAD synthesis under normal conditions. - Understanding cellular pathways holistically is crucial, as isolated findings in vitro may not reflect physiological processes accurately. 24:52 *📊 Limits of NAD Precursors in Humans* - NR supplementation shows an initial increase in NAD levels in humans, but this increase plateaus despite continued dosing. - Excessive NR can lead to increased excretion of methyl nicotinamide, indicating saturation of the salvage pathway. - Balancing methyl donors becomes crucial to avoid methylation issues when supplementing with NAD precursors. 26:31 *🌱 Clinical Implications of Methyl Group Deficiency* - Methyl groups are essential for DNA repair and epigenetic regulation, influencing gene expression and cellular health. - Symptoms of methyl group deficiency can vary widely but often include fatigue, reduced recovery, and immune system challenges. - Maintaining adequate methyl groups supports overall cellular function and health. What are the food Methyl donors? I'm seeing changes... I'm taking Time+ and alternate with DON NMN and TMG. Seeing Hair, nail growth, lack of desire for alcohol, huge appetite increase, deep dreamy sleep. This one is different: I'm a big meditator and noticed this too I'm more able to be in the moment! I'm taking Sert 6 activator in the evening so that may play a part as well. I'm 62 and do HIIT a couple times per week in addition to a walking and body weight exercises. I also do IF and some multiday fasting periodically. Any Food Methyl donors we can eat?

Hi Sean, thanks for the question. According to the biochemistry, the idea would be that you would not need to. The activation of NAMPT should convert the NAM back into NAD and so it would not build up and require methylation to get it excreted.

Hi guys. Another question from me. I'm trying to get an idea how much this new NAD booster increases NAD compared to the 700mg of NMN I take. From the clinical trial it was reported to be 18ng/ml from baseline but that doesn't seem very much. I'm sure I'm missing something. NMN doubles blood NAD levels which I believe is 200ng/ml. Thanks again!

Just supplementing NAD precursors (NR, NMN, nicotinamide) is like pushing on a string. You need to consider the holistic system, which includes the salvage pathway (activating NAMPT, which she covers in the video). Google the NAD salvage pathway and you can find visual representations.

They mention a study in which ALA shorten mice lifespan by a third. It is this one: DOI: 10.3233/JAD-2012-120130 ALA is interested though, hard to know whether to take it or not..
