    This video satisfies what I see as teaching biochems biggest misstep -- missing the forest for the trees. I get so many students that are so dialed in to specifics and miss how much easier it is to see it as a peice of a bigger picture. Nice job.

    Cofactor for PDH are Tender ( thymine = B1), loving ( lipoic acid) care ( CoA = B5) for ( FAD = B2) naD ( NAD = B3 )

    The cahill cycle also gets rid of ammonium ions from muscle cells that has accumulated from muscle metabolism. In the liver the amin groups enter the urea cycle.

    I've seen a few questions regarding LDH deficiency: NAD+ accumulation is seen and leads to inhibition of glycolysis leading to exercise intolerance.

    Wrong Pyruvate accumulates NAD+ is depleted Increased NADH/NAD+ inhibits glycolysis and TCA cycle , and favors lactate production

    Can you, please, explain why in Pyruvate DH deficiency or in B1 def.in alcoholics, pyruvate just does not get converted into oxaloacetate by pyruvate carboxylase to keep the TCA going? 🙄

    I think high NADH/NAD+ ratio in alcoholics favors Pyruvate conversion to Lactate. And also high NADH/NAD+ ratio favors OAA conversion to malate so even if OAA is made from Pyruvate it may be converted to Malate right away. Acetyl coA is available from beta oxidation but there is depleted Oxaloacetate stopping the TCA cycle. Correct me if I'm wrong

    And one more thing, in PDH deficiency the problem is not OAA deficiency but acetyl coA deficiency not getting made from Pyruvate specifically from glucose. To make up for it acetyl coA must be produced from fats. That's why high fat diet and ketogenic diet with low intake of carbohydrates is the treatment for PDH deficiency to make acetyl coA from beta oxidation so as to continue the TCA cycle.

    Mistake that should be corrected The muscles have stored g6p but cannot be converted to glucose that is because muscles lack g6 phosphatase.. So it is g6p that is converted to alanine not glucose. Because if that was the case why would muscles convert its glucose to glucose again. Correct this mistake plz.
