@ninjanerd First off I wanted to say that I love your videos. They've been keeping me afloat through med school. I usually don't comment at all but at 11:50 in the video you talk about codeine in slow metabolizers and in those cases I think it would actually be more beneficial for the children with low metabolism because codeine is a prodrug with the active metabolite causing respiratory depression. So it would actually be ultrametabolizers who are at risk for respiratory depression since they're turnover rate into the active metabolite is increased.

I have a question regarding 18:48 - when turning a pro-drug into an active drug the effect is vice-versa. So an inhibitor such as Omeprazole would become an inducer, no? But then at 24:24 in case of a pro-drug activating we still have Omeprazole being an inhibitor. Not getting my hopes up that someone will see this comment and help out :)

Hey Ninja Nerd, BIg respect for you guys. I have been following you since last few months, and trust me no one has ever made me that focused to learn something as you did. i Dont know if you have time to respond on my confusion but still dropping one question here, So, as you said about codeine, with Poor metabolizer it increases toxicity in blood because it doesn't metabolize. but Codeine is prodrug of morphine so according to that with poor metabolisation it will not change into active drug and will not show the efficacy or therapeutic effect right? it means there are no chances of toxicity. please kindly reply and resolve this confusion. once again thank you so much for all the videos. it really helps. it does.
