Mismatch Repair Animation

Mismatch Repair Animation - This biology lecture explains about the DNA mismatch repair mechanism. DNA polymerase replicated DNA uses proofreading function is to prevent the mis-incorporation of bases however DNA polymerase does incorporate an incorrect base for approximately one in 1 million bases synthesised resulting in a mismatch that distorts the structure of a double helix curiously for fewer than one in 10 to the six mutations actually arise the mutation rate in an E. coli cell is only one for every 10 to the 9/10 to the 10 bases replicated the cell has a number of repair methods including one called methyl mismatch repair to remove and replace of mismatched base methyl mismatch repair involves recognition of the methylation pattern in DNA bases E. coli methylated DNA of the palindromic sequence GAT C using an enzyme called deoxy adenosine place would be another lace them another lace adds a methyl group to the adenine in both strains of the sequence adenine and finding bases pairing normally BMF LA Sax alone but not immediately after replication of the DNA sequence DNA has methylation sequences to panel strand will be methylated but a few stray will still be on methylated the difference in methylation allows the cell to distinguish the correct sequence presumably of the parental strand from the incorrect sequence of a new stray and a series of proteins called mute as new Dell and new age at together to prepare the mismatch named as first binds to the mismatch and recruit me tell you age Mintel recognises the methylated strand and brings it loop may age them cleaves the un-methylated strand containing the mutation view of the GAT C and D a healer case, you the ID unwinds the clean strand exposing it to a variety of exonuclease is the result is a gap that is filled in by DNA polymerase one the DNA polymerase continues through the region that originally contained the mismatch inserting a correct nucleotide this time after awhile the DNA polymerase falls off the DNA and a gap that remains is sealed by DNA ligase the methyl directed mismatch repair proteins and genes are called mute because strains that are defective in one of these proteins have a high mutation rate of bacterial strain with a high mutation rate is called a mutated strain Thank You for watching our videos from Biology Animation Videos channel. I used this "trick" to help memorize what MutS, L, and H were. The S stands for "signal", it signals that there is a nucleotide mismatch. The L stands for "loop" because it forms a loop. I could not come up with anything for the H, but shortly after a helicase called UvrD unwinds the DNA. Hopefully this helps someone.

Please correct the video. It is Pol III filled the gap instead of Pol I.

DNA pol III never helps in repair mechanism. Always DNA pol I in repair mechanism. So what is shown here is absolutely true.

She’s right it’s DNA Pol III
