As a medical student who JUST came out of the basal ganglia lecture for the direct/indirect pathways, you are amazing. This clears all the foggy debris my professor was spewing our way. Can you please teach the rest of my neuro course?

I have an exam about GTS, which is a complex syndrome... and to understand where the tics are coming from - I had to understand the basal ganglia circuit. There are some hypothesis about how the connection between the substantia nigra and subthalamic nuclei isn't working as it should - the substantia nigra should be telling the subthalamic nuclei stop, when it receives too much dopamin - but it seems as if it never does, when a person has GTS. Because when a person has GTS there's an increase of dopamin production and that's where the chemical dysfunction is being expressed. I am so thankful for your help. It's the only way I could understand this.

Hey does anyone know if the Substantia Nigra inhibits the subthalamic nucleus with D2 NT like in the Indirect pathway. or is the substantia nigra using GABA like the rest of the CNS? I'm assuming that the Subthalamic nucleus excites the SNcompacta with glutamate like it would the Globus Pallidus Interna. Thanks!

Marry me Emma, after weeks of frustrating study I found that my lecturer was wrong and so were my peers. I spent days trying to understand something that was wrong with my friends who just ended up rote memorising the pathway.

William McGilberry Hey hope this isn't too late The STN receives input from both the cerebral cortex and the GPe (indirect pathway). The cortex stimulates the STN whereas the GPe inhibits the STN. Also, the SNc receives input from the limbic system and prefrontal cortex. Consequently, it is thought that the SNc plays a role in facilitating movement that seeks the most rewarding outcomes.
