Gram staining for differentiating bacterial species

Gram staining (or Gram's method) is a method of differentiating bacterial species into two large groups (Gram-positive and Gram-negative). The name comes from its inventor, Hans Christian Gram.
Gram staining differentiates bacteria by the chemical and physical properties of their cell walls by detecting peptidoglycan, which is present in a thick layer in Gram positive bacteria.[1] A Gram positive results in a purple/blue color while a Gram negative results in a pink/red color.
The Gram stain is almost always the first step in the identification of a bacterial organism. While Gram staining is a valuable diagnostic tool in both clinical and research settings, not all bacteria can be definitively classified by this technique. This gives rise to Gram-variable and Gram-indeterminate groups as well.

    The platinum loop after red hot should be kept nearer to the flame for few seconds so that it doesn't kills the bacteria which should be taken next for observation/differentiation.Nice informative video.

    The techniques are good. But I think this staff should mark the position of bacteria by drawing a circle on the slide before starting stain them.

    You do not need to let it air dry. You do the heat fix to not only dry it but it kills the bacteria and makes it stick to the slide.

    Sibusiso Sibiya random but did you use the same times as the guy in the video did and succeed? (30 seconds methlyne blue, 1 min iodine, rinse till clear with ethanol, then 1 minute saffranin?) My teacher told us (1 minutes MB, 2-5 minutes of iodine, 6 seconds of ethanol and then 2 minutes of saffranin) and I spent the whole lab yet can only achieve all purple :( so Im now wondering if i put the stains on too long and then the ethanol not enough. 🤷🏻‍♀️

    jennifer pan it usually means you didn’t run the enough ethanol so next time run it till you don’t see any blue color at the bottom and immediately rinse

    Anyways for heat fixing you shouldnt heat fix too long as you might crack the glass slide instead. Instead just move in and out the flame 4-5 times can already.

    Not really 10 seconds, as long as you test it on the agar, it doesnt burn the agar it should be cool enough to extract a bacterial culture

    Yes we have to wait orelse the inoculation loop which was incinerated,it will be hot so when you keep that loop in the bacterial culture there may be chance in which bacteria may die.
