    How many mL of water can one use for serial dilution regardless of bottle/tube size. Can it be whatever ? As the main goal is to dilute and isolate ? Appreciate you

    I find using a total volume of 10 mL for each dilution convenient. Start off with 10 mL, add/dilute/shake, then use 1 mL of that to the next container with 9 mL of new water. Do that 3 times and then you will have your 1X, 10X, 100X, 1000X. I plate out 1 mL of each of those. Then you get a wide range of germination rates to choose from.

    What type of agar recipe are you using? Will the agar absorb all that water???

    Standard MEYA. It will absorb that water in 30 minutes.

    So do these methods to get a diko an mono an then would you take agar transfer an put both in a dish to grow out an hope it's a successful mush cross?

    @@unknowni1190 No. You described what you do. One swab, two fruit. Put it to agar. I never use that technique, but I'm sure someone else has a video on it.
