Can you go into detail for a future video about using microwave, and what all can be done by using a microwave. Example, can we do LC, Agar plates, grain, basically just how much can we do by using a microwave. Thank you for all you do!
If bacteria is eating away at carbon and releasing the scent of ammonia doesn't that imply that there is a bacterial infestation? Still worth keeping the substrate emitting ammonia smell?
The ammonia smell is a new one for me. I have never smelled anything like that. People often interpret things differently, so it might have been more of a fishy smell which I have noticed. The substrate has probably gone anaerobic, which is what many bacteria like. I would throw it out immediately. Not much chance for success if bacteria are in there eating the nitrogen-containing compounds like amino acids and protein.
@edwardgrand I've encountered with pasteurized and sterilized hpoo. Emits a strong pungent oder similar to windex or cat urine. If the root cause of the smell is bacteria, then shouldnt manure get sterilized? But Wayne Myco mentioned that the ammonia smell is most common with sterilized manure but this seems counter intuitive?
That needle and syringe could be reused many more times.
The chance of contamination is not worth a 20 cent needle and syringe.

No. I just waft the air out once a day. Depends on how well your tub seals and ambient temp/rH.
Rh staying at 99% using puppy pads, temp is between 78-80.2F
@edwardgrand ...and I'm using modified 105qt "monos" (they will only be used for this) 4x 6qt SBs fit in a grid. I do have .5in breather hole clusters with cloth tape filtering them, they're located where mono holes usually would be I guess. Edit: you da man!

Man those are pretty.. I heard you say you boil your sub and you use perlite sometimes, is it ok to use perlite in leu of, or with verm in a bucket tek?
I stopped using perlite. Just wanted to get rid of it. I'd stick with the verm or just do straight coir.

I think I've just watched this again for the like.. 10th time, easy! Something I've always been wondering.. can syringes and/or needles used for LC inoculation be PCed and re used? I'm running through quite a lot, they're cheap but it's a lot of plastic in the bin..
Yes, they can be PCed, but they tend to distort and become opaque.
QQQ is LAB what we are achieving by fermenting grain before boiling and sterilization? If so wouldn't that eliminate the need for Dr mics? I know when I ferment peppers in a salt/sugar water mixture that is the beneficial component. But sterilization would kill it, therefore leaving it now nutrient rich food for mycelium? I have no idea if I'm even close here, can you shed more light on the matter
Yes, PC would kill any LAB. I'm not really sure what the 'fermentation' does. I think the idea is to start breaking down some of the polysaccharides in the substrate so that they become more readily available to the fungi.

@@matteocicchetti2086 yes, AC will dramatically lower the relative humidity. We are going to do a live in about 3 hours.

Great video! So can I throw my shoeboxes without lids into the large tub as soon as I make them up or do I need to wait on them to colonize with their little lids on and then remove them later?
I usually wait 5-7 days before I remove the lid and put them in the monotub.
@edwardgrand cool deal.. I have several for my first run and i put them straight to mono's and it seems they've stalled. Also, I saw where you told someone the temp needs to be around 80f.. my temps were around 74.5-77 with 100% rh. Did I mess up? The rhizo is shooting up like zombie hands but not consolidating very quick. One set was spawned on the 27th, another the 29th and the newest on the 4th.. I've adjusted my temps, should I just take the boxes out for a few days?
So do these methods to get a diko an mono an then would you take agar transfer an put both in a dish to grow out an hope it's a successful mush cross?

I've been messing with all kinds of recipes.
Wouldn’t that contaminate the spore if the agar was bad? Id scrape spores onto agar.
I usually pour my plates a week before using them. No reason they would be contaminated. The idea is to separate the spores from possible contams on the print by 'streaking' them across the plate.

I prefer agar for everything. LC is fun to play with, but mostly for mass production of one cultigen.
I actually just barely got my plates on point.. tried going with some green agar bars I found locally but didn't workout 2 good finally I ordered a pound of agar powder online n can't stop me now ... I Tell You What!
