Gene Silencing mechanism

Gene Silencing - This lecture explains about the gene silencing mechanism by antisense RNA technology. Antisense RNA and RNA interference mechanisms are used to control the expression of desired genes in cell and it can be used to degrade target RNA so that the expression of proteins can be prevented.

    So many errors are there in this video...RNase H degrade RNA in an RNA-DNA hybrid. In this case an RNase L has been cited to act rather than RNase H. Now a days it is said to reside in the Dicer.

    you are saying that if SOD 1 is mutated or fails to function then it may leads to ALS due to reactive oxygen species which mainly affect brain cells. Why there is need to silence this gene. Suppose if u have already ROS in our body it may affect brain cells which also leads to ALS. How is it possible sir???Please explain.

    RNAi or the double stranded RNA technology does not degrade the DNA, it infact degrades the specific mRNA against which the antisense RNA or the double stranded RNA has been incorporated in the cell. But in this lecture it is being said many a times that the DNA is degraded. which is completely incorrect.

    If you disrupt a gene (by inserting a transgene), then observe what was the effect on the phenotype, you'll identify the gene's function. How to identify the gene you've disrupted? Simple. Use a probe for the transgene you inserted & isolate the segment. The sequences on both sides of the transgene will be the gene disrupted. This is known as gene tagging.
