Drosophila melanogaster: Handling | | UPV

Título: Drosophila melanogaster: Handling
Descripción: This learning object describes the main characteristics of Drosophila melanogaster and you will learn how to handle this insect for its observation in the laboratory.
Descripción automática: In this video, the presenter provides an overview of Drosophila melanogaster, commonly known as the fruit fly, which is used for research in laboratories. The video explores the species' characteristics, such as its short life cycle, four pairs of chromosomes, and various mutant types, providing visual aids like pictures of the fruit flies with different eye colors and body mutations.
The presenter details the stages of the Drosophila life cycle and discusses the practical aspects of handling them in a research setting. This includes the materials and methodologies necessary for maintaining and studying fruit flies, such as using a magnifying lens to observe them closely, methods to anesthetize them with CO2 or cold, and techniques for transferring and sorting the flies without harming them or contaminating the culture.
Further, the video demonstrates the process of transferring the anesthetized flies between containers while highlighting the importance of quick and careful handling to prevent escape or injury to the flies. The presenter also stresses the importance of proper storage and immediate resealing of containers to avoid contamination and volatility issues.
Finally, the presenter concludes the presentation with cautionary notes on the handling of fruit flies to prevent their escape or death, while also mentioning references for further study into the subject of Drosophila melanogaster research.

I have a science fair project, and I will be using Drosophila. This video was very helpful, because I will be doing the experiment on my own. Thank you!

Respuesta del profesor: Thank you for your question. There are several medium for Drosophila, some commercial, and some that you can prepare yourself. The medium that we use in the laboratory include flour, yeast, water, agar and sugar. Also, you can add potassium sorbate, propionic acid and sodium nipagin to control fungi and bacteria. We don't use banana pulp.

The teacher answers: "Thank you for your question, but we do not use glycerin to observe the sex comb. In adult stage we just need the lens. In pupa stage we can use water to observe them better if it is necessary and also to maintain the pupa wet and avoid to damage them".
