Dear sir What is the role of active and deactivate OH group in C8 and C18
So retention time and separation depends on polarity of molecule in some cases retention time less for c8 column and some cases in C18 columns . It depends on mobile phase and analyte polarity

C8 column hydrophobicity lesser than C18 column , when a polar molecule will retain more time on C8 column than C18 column . So retention time less in C18 column than C8 column .

Can you use C18 or C8 columns be used to separate mixtures containing amine salts. I.e. will it be able to separate a mixture of for instance serotonin.HCl and dopamine.HCl ?

@@dipuswapnu minimum requirment is NLT 2000. If more than 2000 than it is very good bcoz peak sharpness becomes better when your theoritical plates are increased...............
