Deflasking the "Rarest" Aroid from Tissue Culture | Philodendron Spiritus Sancti

G'day, guys! In today's video, I deflask some Philodendron erubescens 'Red Anderson', El Guapo, and the "rarest" aroid indoor plant out there, the Philodendron Spiritus Sancti. Connect with me on Instagram - / petevsplant

Deflasking tissue culture plants may seem complicated and daunting, but I assure you, it's not that hard at all. Today, I show you how to put together the pots, the substrate (coco coir and perlite), and the process of sterilising the tissue culture plantlets before planting them up. This is how to deflask the 'rarest' indoor aroid plant Philodendron Spiritus Sancti.

    Roots rotting off makes sense: agar is constantly moist of sorts. They need a different type of roots when transitioning to soil or water.

    Thanks for the awesome videos Pete! I'm working on some of my first tissue cultures and your videos have helped a lot for them and also my many other aroids and such. I started acclimating my first batch a few weeks ago and now have 3 more different kinds working on acclimating out of the agar now. So far so good.     @@petevsplants7516 that's great to hear! My WW look to be getting ready to start growing. A couple have some beautiful variegation. The others are fresh out of the agar one week ago. So they will probably take a little while yet before I notice anything exciting. The Peraiso Verde also have awesome colors. I'm definitely glad I ventured into the TC's

    Bruv I myself would love to get into TC plants, it does seem like a little work but well worth it in the end. I almost bought a bunch of TC Joepii plants to grow out but I decided I didn't have the gear to work with them but then I met a fellow in Washington and he did it in his room and sells some amazing little plants.

    Aloha from Maui Pete, Well Done!!!!! **note: on prop boxes, or “Hotel de la Totes”, I heat a metal poker or extra large nail, hold with pliers & heat tip with gas torch or gas flame, & poke 3 or 4 holes per each side of tote. ***drilling seems to catch & crack plastic tote*** Humidity will stay extremely high, but not excessive, & less chance of plant leaf rot…..     Nice work cheers for sharing. On the plastic clear cup thing, its rather ridiculous right especially when the biggest polluter is the likes of the plastic coke bottles and packaging etc in the super markets. Crazy world we are living in

    I think it could work, but they'd have to be big enough to fit in those rubber inserts properly so the roots could get access to the water/air below

    If you’re looking for disposable plastics, try the $2 dollar stores in your area. They usually hold onto those sorts of things. There’s also been floating around in Daiso (japanese $2 store, not sure where in Geelong but def in the CBD and some bigger shopping centres) these plastic pudding cups that make a perfect humidity dome and base! Hopefully that helps.

    I’ve been recycling Starbucks cups for my propagations, they’re a perfect wide mouth with a lid, plus recycling. Plastic is definitely disappearing (thankfully!)

    Spiritus grow like weeds. I have one in a 4in/10cm pot with roughly 7in/18cm leaves & it gives me a new leaf every month. even the aerial roots grow quickly so I’m not surprised that your plantlets are already growing aerials

    Cool vid Pete. Kinda surprised you didn't throw a few of these into your "turbo" propagator system. Seem like you'll get the most amount of roots quickly that way. Also would using rooting gel help or is there enough hormone from agar cocktail?

    Yeah, ideally you'd probably do that too by baking it. Though, I've never had a massive problem (in my limited experience) from the substrate assuming it's not soil. If it were soil, then you'd probably want to definitely sterilise it.

    Defs recommend inoculating with mycorrhizae, something broad spectrum after deflasking. At least from my experience with orchids.

    Interesting. I guess you have to do it in the soil and not include a fungicide after you've soaked the plants or it may defeat the purpose.

    Hi Pete, I'm a big fan of your videos and watch them whenever you release them. I have a question what's the point of using hydrogen peroxide immediately after removing from the sterile flask? Isn't it an extra step? Don't you think that might negatively affect the new roots? When deflasking orchids we just wash it in water and fungicide could be sprayed later on or same way you're doing it.😊

    I honestly haven't tried it yet haha I bought the stuff but just haven't had time. Recently, I found coco coir can be a mixed bag, because it lacks nutes if your plants are hungry and growing fast, you have to fertlise or move to soil quickly which can be a pain.

    Heya Pete, tissue culture plants are sterile, so I’m curious about why you’re hitting them with peroxide. The copper makes sense as a prophylactic if you don’t rinse it off.

    The plants from a flask are already sterile when you take them out, so you don't need to sterilize them again. Just wash off the gel and slowly acclimatize the plantlets to your growing conditions.

    I always get so jealous of the plants you get haha, i buy some new ones every few months so i don't give my husband a great attack 😂😂😅. I wait until they drop a little in price before getting em. I love the spiritus sancti, but its to much for me right now, same with the el guapo, so beautiful. But maybe next year im thinking. I yried to buy tissue culture white wizards, and they all died, i wasnt to upset, i was out like 60 bucks. I just went ahead and bought a acclimated plant. I didnt give up though, i have a beautiful highly varigated thai constellation, and a milk confetti that im trying again on. So far, so good, they've lasted just over the week mark so far. If you would ship to the USA, please let me know, I'd pay for the shipping. 😅😅😅😂😂😂

    Oh nice work! Yeah, I feel you re: price of some of these things... I wish I could ship outside Aus but the laws are such a pain here that it's not worth it.

    Shit load of bleach, literally! 😂 Leaving ANY agar scares me. I lost 7 of 10 aglionima pictums tc from the stems rotting off. I fully attribute it to not getting off all the residue. My tc TC is doing fabulous! I was super anal about washing off 100% of the agar. Even used a baby toothbrush on the roots. I also inoculated the roots with mychcrorrizal and azospirillum. I think dipping in copper and not rinsing makes sense. I’d be going for the residual of the fungicide. If you rinse, you kinda did the exact same thing the hydrogen peroxide bath did. Congratulations on the PSS! It would have taken me at least 30 min each cleaning those bad boys up. Especially as rare as they are in your area. Locally, the prices have pretty much collapsed to reasonable. You can get small rooted plants for ~$250. Seeing tc for double digits and even seeds….but…? I did do a thing though! Rooted node with a sprouting leaf BMF inbound soon. Couldn’t pass up the chance @$125 shipped. I enjoy growing lil things into big things and I should be way ahead of the game vs waiting for the price to continue to drop. (Even if that’s not true, that’s my story and I’m sticking to it!)

    Yeah that's crazy re: prices in the US. BMF are still $2000-2500 per leaf for an established plant and a large PSS would be $5k+. TC should bring them down but it'll probably take another year or so. Everything's 1-2 years behind the rest of the world here in Aus because of our importation laws.

    @@petevsplants7516 Just watched the unboxing video North Shore Tropicals did. Vancouver, Ca is about two hours north of me. Those small PSS and BMF she unboxed were priced at $228US. Super solid prices, especially for a nursery.

    That's ridiculous... as I said in the comments, each of those would've been 10-20x that price in Aus. I saw a 1 leaf BMF the other day and the price was $2500 AUD

    A bit of acidity is good for your skin. I never had any problems with 3% hydrogen peroxide. If it foes not hurt leaves and roots.... In the end it's just water and oxygen.

    I think the reason it doesn't harm plants is because of their cell walls, but with other organisms it destroys the cell membranes.

    No personal experience, but I see a lot of people rooting pups and qorms in fluval stratum very succesfully. No root battles like with sphagnum and both are pretty sterile. I worry about emerging roots staying moist enough in your soil mix. Vermiculite is known for developping roots and seedlings. Another option?

    Fluval stratum is my go to now for props. I’ve not tried TC yet though. It’s also great mixed equal parts with perlite and moss in prop boxes - the moss doesn’t bond so tightly with the roots. MST comes off with a gentle shake but the stratum and perlite chunks make it super airy and loose. Worth mentioning though that the stratum /perlite/moss mix looks a sight due to the blackness being absorbed by the moss. Corns root and sprout leaves in 2-3 weeks. My various wet sticks activate far faster than with pure moss prop boxes. My prop boxes just get window sill light not grow lights.

    I have been wanting to buy some tissue culture plants, but it’s so hard to know who is a trustworthy seller. Anyone with any recommendations? I am in the US.

    I think you just have to try different places until you find someone reliable. That's what I've done at least.

    Haha no... those are about $20k Australian still. Definitely not in my price range.

    Yeah, they're in TC but there're clearly not many of them around as the prices are still so insane.
