Sir is talking about force applied and in F=- kx ,F is restoring force of spring because restoring force perform in opposite direction of ist displacement of spring so here is a negative sign

That’s the force of the spring. The force is equal to the negative spring constant multiplied by displacement. The ENERGY however is half of the spring constant multiples by the displacement squared

Yes but not from hand from our muscles we apply force on string which increases as we go on compressing more(as more distance is covered )
