Bioprinting 101: How to make Microfluidic Chips

Student In STEM Ria Bhatia demonstrates how you can create a pH gradient generator chip using microfluidics. Through this experiment, you will familiarize yourself with TinkerCad, a widely used 3D modelling software. Additionally, you will learn about microfluidics, which deal with the flow of liquids inside micrometer-sized channels, and receive an introduction to flow concepts, such as laminar flow. Throughout this project, you will also learn about acid-base reactions, which are indicators of pH value, and technical skills, such as ImageJ and Excel analysis.

Can i have more info about the dimensions of the channels?
Is it advisable to use a vacuum pump in order to quickly remove the air bubbles in the PDMS?

cool! how many more times can the printed design be casted? or doesit disinttegrate aftyter first cast?

It's one print per cast. Once you cast, it solidifies over time. Then you can wash off the channels.

Really interesting
Man, I'm just trying to figure out how colors mix to produce different colors, like is it linked to turbulance and if so is it a chemical reaction, or an optical illusion.

The color is based on the pH of the fluid. I think they used one universal pH indicator for both samples. So, when they change color, they change due to a chemical reaction.

Are the channels exposed to the open air on the bottom face?

Yes we use tape to cover the channels so you have a seal to flow fluids through. Otherwise you can use a glass slide.

Any recommendations on how to attach the PDMS device onto a glass slide ? Any help is much appreciated

Just make sure both surfaces are clean and dry before attaching the glass slide onto the PDMS cast. Good luck!

You have to go for O2 plasma etching of the glass slide after which you bond it with the PDMS

why dont they just completly 3D print the end product in plastic? with channels built in?

depending on the kind, but fdm is out because of the porosity , i don't know for resin printing but i guess you'd need something chemically inert , as for slm/sls, it's probably feasible yet would be an expensive piece of equipment
