Sue Perkins was originally pencilled in to appear with Greg. She was mentioned as appearing in the Christmas TV guides such as the Radio Times, but had to pull out at the 11th hour for reasons unknown and was replaced by Richard Ayoade. No disrespect to Sue, but I don't think "𝙱𝚊𝚍 𝙳𝚘𝚗𝚐" would have gone as far as it did had she been there.

There's an episode of Qi where they cover this by showing a multi-colored three-dimensional cube which is partially and what is represented to be shadow but two different squares look very different colors even though they're actually the same exact shade but the illusion of light and shadow makes it look different when our brains try to interpret the information

I saw him live and could tell he was kinda suppressing it because the couple times he let out a honk everyone laughed. Not great for timing and delivery. Jimmy Carr isn't famous because of a strange laugh. He's a genius joke writer. Wouldn't need something like that. Also, a weird laugh is a curse not a blessing and if you could NOT have it there are way more benefits than the gain from faking it. THE END

That's not a fake laugh, he's always laughed like that. There's a random short video clip of his sister who laughs the same way. Apparently it might be somewhat genetic because their mother also had a weird issue when she laughed it would cause her to black out.

It was a joke. Whether you find it funny or not is up to you because humor is subjective. Was probably staged because there is no way this is broadcast live, so obviously if it's pre-recorded and they are taking breaks, they could easily just keep that extra recording of when they were goofing off on the break before the break ended.
