just got out of COVID quarantine and my prof didn't explain this at all and the figures were making it really confusing. Can you explain why did you put carboxyl group in ionized state, whereas amino group was not ionized.(in cell conditions both are ionized)

For Glycine when pH is 12 how does the net charge is -1? In N-H because of the deprotonation the charge is -1 and in COO- bond there is -1 charge so the total net charge is supposed to be -2. When pH=7 you are counting the charge of COO- is -1 to get the total net charge. Then why when pH=12 you are not counting the charge of COO- group -1 charge to get the total net charge? Can you please explain it?

Where can I find the second video
Sir, I'm a JEE aspirant and I recently faced a problem on Biomolecules The question had three phases of glycine where [A]- Both acid part and -NH part was protonated [B]- Only the -NH part was protonated [C]- Both the parts was deprotonated pKa1=2.3 pKa2=9.7 The question asked if at pH=2.3, half of [A] is converted to [B] or not? So sir, can I consider it to be correct that yes it is converted, on the pretext that half part is protonated and half is deprotonated? (I am having confusion in the part where it asked 'converted', and I am not sure that half protonation would result in half conversion of phase [A] to phase [B])

In case of titration of a peptide chain, should we start from the fully protonated form of the peptide or the fully deprotonated form?
Where I can find the second video for polypeptide?
is net charge would be -2 at pH 12 since there is already -1 (pH 12 does not change -1 and -1 stays -1) in carbonyl group and very basic condition(pH 12) rips out H from NH which makes -1 as you mentioned, So -1 + -1 =-2 isn`t it?

The pI of glycine is written as 5.97 in books. Is that correct if we take ph 5.97 instead of 7 in this calculation? In books the formula for pI is , 1/2 (pK1+ pK2) i.e., 1/2 (2.34+ 9.6) = 5.97

Are you sure that the pI for glycine is 7 and not 5.97...🤔🤔 Isn't pI supposed to be calculated as (pKa1+pKa2)/2 For glycine it would be (2.34+9.60)/2=5.97

I don't get why the net charge of pH 12 is -1. Can someone explain why? I thought it would be -2, this is because there would be a negative charge from both the nitrogen and oxygen so (-1+-1=2). I don't understand why he ignored the charge in the oxygen in pH 12, but accounted the oxygen's charge in pH of 7.
