Draw your character in illustrator > drag and drop your ai file into after effects > place the asset into the Timeline > right click the layer > create > make paths. Can't imagine character drawing in AE ever be easy unless you're going for smt super easy.

Do I necessarily require a stylus for digital art. Can't it be done by a mouse.

Yup can be absolutely done with a mouse using the pen tool or shapes and creating points and morphing them

Which are the graphic designing softwares which Don't require stylus.

softwares don't require you to use a stylus.. you choose to use it to make a task easier.. of course you can use your mouse if you are comfortable with it
