Lysergic acid diethylamide, or LSD, is a potent psychoactive substance that is commonly classified as a hallucinogen or psychedelic drug. In this video, I discuss our understanding of LSD's effects on the brain.

Lysergic acid diethylamide, or LSD, is a potent psychoactive substance that is commonly classified as a hallucinogen or psychedelic drug. The effects of LSD vary substantially depending on the dose, the mood of the individual taking the drug, and the environment the drug is taken in. Some common effects include mental and physical stimulation, perceptual distortions, and generally positive mood states.
Although we have some understanding of LSD’s activity in the brain, it is not fully understood how this activity leads to the subjective experiences people have while taking the drug. Regardless, it is thought that LSD’s activity at a subtype of serotonin receptor known as the 5-HT2A receptor is critical to the drug’s psychedelic effects. LSD acts as a partial agonist at the 5-HT2A receptor, which means that it binds to the 5-HT2A receptor and generates a response that is a fraction of what the natural ligand, serotonin, generates. LSD also interacts with other receptors, although the role of these receptors in the effects of LSD is less clear. For example, LSD binds to other serotonin receptor subtypes as well as some subtypes of dopamine and adrenergic receptors. Research suggests that one large-scale effect of LSD on the brain is increased glutamate transmission in the frontal cortex, which is thought to play a role in the effects of LSD.
Repeated administration of LSD over several days promotes rapid tolerance, where the drug has much less of an effect than it did initially. This tolerance is thought to be associated with the down-regulation, or decrease in number, of 5-HT2A serotonin receptors. LSD is not, however, considered to be addictive. And, although it is a potent drug, it is generally considered to be non-toxic and safe in terms of its effects on the body.

Be careful. It helped me too overall but you know the rumor about like people thinking they r a glad of orange juice and then never recovering. I got very close to that in 2020. I stayed away for a week 04/28/20 - 05/08/20 and I didn’t come back to reality until July 3rd. I had taken a gel tab . Just one. I puked 30min after I took it and I could tell it was gonna be a wild ride. It was. I had full psychosis. I’m recovered now. Fully recovered I have a job. Been in a three year relationship. Found real God . (I know the spiritual aspect if very all connected which is still true it’s just that Jesus is the true God) idk anyways… Be careful!!! I still smoke pot. I take lithium now too it’s a medicine.

I’ve tried LSD a few times with mostly positive experiences. However I had a very bad trip a few days ago. Mine lasted over 24 hours of sheer paranoia and fear, the likes of which I wouldn’t wish on my worst enemy. My body was completely numb and I felt the extreme need to want to hurt myself and others. I somehow managed to calm down by drinking herbal tea, listening to soft music and talking with a suicidal hotline. If you ever try this drug, make sure you start with a very low dosage, have zero responsibilities and have a trip-sitter with you for at least 8 hours.

this is the main problem with LSD, one bad trip and it can fuck you up in the head quite badly, it happened to me as well and I think I still have "ghosts" of that bad trip in my personality, looking forward for healing myself this year.

I disagree,, personally i prefer LSD over mushrooms.. to me the best description i can come up with is LSD is a cleaner trip Mushrooms are a heavy body trip and makes me nautious sometimes and i yawn alot on mushrooms and feel lethargic where LSD i am energized and alot more happy

@@danparts9333 I personally find that the comedown from LSD can be uncomfortable while mushrooms generally ease you back into sobriety. I think most of the discomfort from LSD comes from the vasoconstriction which leads to you feeling cold and clammy.

It has helped me develop more cognitive perspectives of other people and develop more sympathy for people. I was kinda self centered back then. After those times, I felt like using the drug would be pointless.

Are there any negative implications of a downregulation of the 5-HTP2A receptor on one’s overall mood or baseline or anything else? I wonder this in the case of microdosers who dose small amounts every 2-3 days.

That's a good question. There has been some speculation that 5-HT2A downregulation might be linked to therapeutic effects, but it's just speculation at this point as there's not much data on it. The mechanism of microdosing (as well as the effectiveness) is still pretty unclear.

In my few years of experience I haven't had any negative experience with downregulation. In fact, after a trip the downregulation takes place in much greater fashion than with microdosing. Some suggest that receptor downregulation caused by microdosing is minimal to none. In high (trip) doses it definitely takes place and after trips most report feeling much better and their psychological condition improve drastically. After my first trip I felt great for the next few months, better than ever in my life before. I don't think it's caused by downregulation though. I think this effect is just a 'side effect', that doesn't have to do anything with the actual therapeutic psychedelic effects.

What is the major differences between SSRI serotonin activation compared to LSD? Why LSD can induce hallucination with serotonin receptors activation, but SSRI did not? Is SSRI cannot activated 5ht2a receptors or there is other major factors that cause these differences.

One thing is that the mechanism is different; SSRIs are inhibiting the reuptake of serotonin, not binding to serotonin receptors. But the question still remains of why the increased serotonin levels induced by SSRIs don't elicit similar effects to LSD. The answer is that no one is sure yet, but one hypothesis is that LSD prompts some change in the conformation of the serotonin receptor that influences how the receptor interacts with other cellular constituents. According to this idea, there's something unique about the structure of LSD that causes the serotonin receptor to respond differently to it than it would to the increases in endogenous serotonin levels that SSRIs would cause.

A key factor is also the much greater binding affinity of LSD to the receptor. LSD is preferred over serotonin itself, the natural ligand! The receptor prefers LSD, if you like.

Also, there are two different pathways activated by the receptor, the G protein pathway and beta arrestin. Serotonin activates the g pathway more, LSD the beta arrestin. Forgive my shocking spelling.

Does lsd affect people in depression? LSD is a partial agonist on 5-HT2A? I doubt this, LSD has strong affinity to it, which makes it an agonist. But LSD has partial agonism on a dopamine receptor, I think it's D3.

LSD has high binding affinity for 5-HT2A, but lower activation efficacy than other 5-HT2A ligands. I relied on a 2016 study (see reference below) for information on affinity, potency & efficacy. From that study: "LSD and psilocin were 5-HT2A receptor partial agonists, with 28% and 16% activation efficacy, respectively, whereas all of the other compounds presented higher 5-HT2A receptor activation efficacies..." Because even though we don't understand exactly what it does in the brain to produce its effects, we do know from animal research and about 80 years of human use that it isn't linked to life-threatening complications.

Bro it's one of the hardest drugs to synthieze and the precursors are really hard to get on large quantities
it may not have physical effect but it has mental effect which is unpredictable and varies from person to person.Don’t get fucked up and loose your sense of morality just to be cool. :)

Yea this doesn't happen unless ur wreckless with it bud. Do ur research on dosage and set and setting. Ur environment controls ur trip and ur state of mind. These drugs have also been proven to be the best treatment for depression and addiction to other substances. No one has ever gone crazy bc of LSD and no one has ever OD'd on it bc its practically impossible to do so

The benefits are only temporary.
Did LSD one time and developed a condition called HPPD. Wouldnt wish this condition on my worst enemy. For those of you who think lsd is safe you may wanna rethink tripping in the future. Quit while you're ahead.
