Things are much simpler these days. There are many unnecessary steps and techniques included in this video that are not currently needed, and the ultramicrotome has advanced drastically.

i dont think that it is nessesary to make marks for trimming becuase anyway you are not cutting on the mark

Block too far out of the holder. Dreadful score marks on the sections - throw that knife away ! Fit a new one or use a diamond knife

I'm glad you did! I started doing ultramicrotome in 1966 using a completely manual Cambridge ultramicrotome, with glass knives, cutting phloem tissue. Very difficult! I moved to the Institute of Ophthalmology in 1971 and finally ended up using a Leica ! I retired in 2011,after seeing many changes in TEM and SEM Happy cutting !

How small is 70nm just curious

Smaller than the diameter of the coronavirus. Coronavirus is approx. 100-120nm
