    @John-of6ch No problem. For mushrooms, ultrasonic extraction is better suited than soxhlet. As explained in the video, in Soxhlet the extracted compounds are exposed to heat in the flask for a long time. That is not good for heat sensitive compounds, because it will cause heat degradation. So you are destroying what you just extracted. With probe sonication on the other hand, you can - break cell walls - Soxhlet does not, and - extract soluble and in soluble compounds - Soxhlet does not extract insoluble compounds Most importantly, ultrasonic probe extraction produces great yields and works at low temperatures, such as -20degC.     @trevorhilditch8890 you could just heat and boil off the ethanol, but don’t! Use a vacuum pump at mild temperatures to preserve heat sensitive materials. A rotovap would be the best.     @quinsongumbs71 Yes you can. The advantage of ultrasonic transesterification is the reduction in excess methanol due to the sono-chemical activation of the reaction/catalysis. Therefore you need less evaporation capacity than in a non-ultrasonic reaction. More importantly you need significantly less energy (vacuum and heat) as you need to evaporate significantly less excess methanol. We are looking forward to working with you on your project.     @jovan9724 The 14mm probe has a higher amplitude than the 22mm. The amplitude of the 22mm probe can be enough for nano-emulsions, depending on your formulation. For larger batches, we would recommend using a flow cell in combination with the 14mm probe. By recirculating or running multiple passages through the flow cell, you will get a better result.     Not only is it extracting the oils, it's emulsifying it. You're used to seeing the oils floating to the top of your brew rather than evenly dispersed as colloidal micro droplets, scattering the light differently. I've seen this look in sonicated vinaigrettes, too.

    Hello. I would like to ask. If I would want to extract mushrooms, is it better to use soxhlet extraction setup or sonicator? With soxhlet to make water extracts I would need to boil water to get water soluble compounds. With sonicator it is only possible to extract alcohol soluble compounds and I will need to make boiling after also to get water soluble compounds if I want double extraction? Or sonicator can not only make alcohol extraction but water extraction in cold water without boiling too?

For the preservation of heat-sensitive materials, it is advisable to implement a strong vacuum and low temperatures during this process step.     @ameendavid4037 Absolutely. After sonication, use a very fine filter paper (e.g. 1µm) to remove all suspended solids. Then use rotary evaporation at very mild conditions. This may take longer but it will give you a better quality. You can use fresh Rosemary. Any moisture of the fresh material will mix into the liquid phase and be separated during evaporation. Vary the water/solvent ratio, the solvent used, the temperature during sonication and the sonication intensity (amplitude) and duration to optimize your extract quality and yield.     @@HielscherUltrasonics I don't think that's really a fair comparison in this case. If you extract mushrooms in alcohol you will extract much more leaving them to soak in 6 weeks then one week with no added intensity

    Comparing the cleaning bath to the UP100H probe sonicator is like pitting a kitchen knife against a heavy-duty chainsaw for tree cutting - it's just not a fair match. The cleaning bath simply isn't designed for botanical extraction tasks, especially not for dealing with the robust Chitin-based cell structures found in mushrooms. The cleaning bath is well suitable for cleaning jewelry or even carburetor nozzles, but when it comes to extracting substances from materials like mushrooms, it falls short due to its very limited intensity. Even giving it an extra 30 seconds of sonication time doesn't bridge the gap in effectiveness compared to the UP100H probe sonicator.     @momsmushroomsjodyfoster5786 There are two aspects: 1) The solvent volume should be high enough so that the flask does not run dry when the solvent evaporates and fills the Soxhlet well. The solvent volume should be low enough to allow evaporation. Our suggestion: At the start, fill the Soxhlet well manually with solvent to just below the apex of the siphon and fill the flask to the level of its maximum diameter. 2) Depending on the compound amount to be extracted, you should not oversaturate the solvent in the flask, which would cause separation (e.g. precipitation). If you see oversaturation, use less botanical material in the Soxhlet well. Have you considered ultrasonic extraction as an alternative or add-on to Soxhlet extraction, yet?     Thank you so much for your fast response! It’s difficult finding answers for my questions! But, again please. Should I be adding 250mills of liquid to the a 500mill flask? Because the liquid expands as its heated, correct? I tried adding more than 250 mils of water, and it was disastrous.

    @momsmushroomsjodyfoster5786 Fill the flask to just above the widest diameter. Depending on the volume of your Soxhlet apparatus, you may have to add a little more, if you see the level in the flask falling too low. Make sure that your condenser condenses all water vapor, so that you don't lose water during the process. You see that our Allihn condenser has a very narrow opening on the top to avoid vapors exiting instead of condensing. Use a low coolant temperature on the condenser. The expansion of liquid due to heat is insignificant. Use boiling stones to get smoother boiling in the flask and use medium heat to start slow. Once you have everything under control, you can increase heat in small steps to speed up the process. Alternatively use cold water and sonication and avoid the Soxhlet all-together. In a Soxhlet extraction, the extracted compounds are exposed to heat in the flask for a long time, causing heat degradation. With our sonicators, you can extract at cold temperatures, so that the compounds are not exposed to heat.

    @momsmushroomsjodyfoster5786 Water can be used as a solvent in a Soxhlet extractor, but it will extract water soluble compounds only. Without sonication, the water or solvent will not break cell walls. Therefore, extraction is very slow and incomplete.     @aravindg54, Yes, you will need a methanol recovery system (heat + vacuum) to separate excess methanol from the biodiesel.     @p_peja Ultrasonic extraction is the most efficient extraction method. You can adjust the sonication intensity on our Hielscher sonicators, to be just intense enough to break the cell structure. In combination with low or ultra low temperatures, this preserves the most mushroom compounds. Thermal extraction on the other hand is less effective in breaking the cell structure and the heat degrades many valuable compounds. Thanks for watching!     @alexmaster9437 As any other mechanical process tool, the sonotrode (horn, probe) is subject to wear and tear. The amount of wear depends on many factors, such as sonication intensity and sonic energy per volume sonicated. Our sonotrodes are made of a high quality grade 5 Titanium. The overall wear into the process liquid is typically in the PPM (parts per million) range. Our customers include food, beverage and pharmaceutical manufacturers, that test their products for safety.     can you set the specific frequency that you want on these machines? I am trying to test how Ultrasonic frequency and duration affects a specific fluid sample. for my application, the frequency range is 20kHz to 100kHz and I am also interested in also knowing the Power delivered. how well are these devices suited for my purpose and do you have any recommendations? I need a device to apply the ultrasound at specific frequencies and a set up to tell me the power delivered

    @raechardbernard We make resonant probe-type sonicators. The resonance is determined by the mechanical design. The generator adjusts the driving frequency to this resonance. This is like a pendulum, that has a single frequency dependent on its length. On the other hand, 20-100kHz is all low frequency ultrasonics. We make various devices with distinctive frequencies from 18 to 60 kHz (higher for some specific applications). Frequency (in low frequency ultrasound range), is not a significant influencing factor for liquid processes. Amplitude and liquid pressure are the most important factors. You can adjust the amplitude in a wide range on our sonicators. Our devices work a various liquid pressures.     @Gta_Selection: We own a Microtrac Nanoflex DLS, a MicroTrac S3500 LD, MicroTrac S3000 LD and a Nanotrac Wave II DLS for particle size measurements and stability measurements. The Dynamic Light Scattering (DLS) systems have an impressive measurement range of 0.3 nanometers to 10 micrometers. So yes, we measured the nano-emulsion in our technical lab, using the Microtrac Nanoflex DLS in this case. We have many customers, that measure their own emulsions on identical or other measurement systems and cross-reference the measurements with our measurements.     @Gta_Selection You should contact StuphCorp (www.stuphcorp.com) regarding the Stuph emulsifier range. Their emulsifiers are food safe. Please understand, that we don't sell StuphCorp emulsifiers. We like their products though. We do recommend StuphCorp to almost all of our clients for CBD & THC nano-emulsions, because it works. We cannot say the same about many other proprietary or generic emulsifiers and stabilizers in the market. We tested many emulsifiers - lecithin and maltodextrin included. Clients bring all kinds of emulsifiers to our lab. When it comes to stability, flavor and price/performance, nothing we tested comes even close. Life is too short to waste time and money on stuff that doesn't work.

    @@HielscherUltrasonics yeah without an emulsifier it won't work, you can use a $30 blender with emulsifiers no need for expensive equipment to bond oil and water, but at high concentration it comes out cloudy the more oil you use

    @anthonyackerman5556 The difference between probe type sonication and an ultrasonic cleaning bath is the intensity - the power coupled per surface area. Due the technical design of an ultrasonic bath, the amplitude - this is the mechanical displacement of the surface in contact with the liquid - is limited to approx. 1 micron. Hence, a cleaning tank couples the power across a large surface area at very low intensity. Ultrasonic probes have adjustable amplitudes of up to 100 micron or more. This couples the power using a very small surface at a much higher intensity. The intensity of an ultrasonic bath is much too low to effectively break chitin and cell wall structures.     @Abricos4440 Our sonicators are great for mixing materials and for making stable emulsions. Hence, our sonicators are not a good tool to separate oil from water.
