  Is it possible for media ( L15 +F12) from a flask to be changed in yellow at 39 Celsius degrees in just the first 20 hours from preparation? Or this change is because of the stressed cells from the media or other unidentified factors ?

  Thanks for sharing the proper method. I'm using the multi-channel pipette, but suddenly I couldn't eject the tips. Instead, the pipette itself is separated into two parts; the handle and the rest. I think the rotating part has something wrong, but I don't know how to fix it. Would you give me a tip for me, please?

  Hell yes. Positive electronic pipettes are insanely precise. I did some measurements, deviations of volume on 1.6% max volume (! 20uL using a 1.25mL pipette tip) are well below 0.007mL / 0.4%. Meaning you can pipette volumes to not even use volumetric glassware (e.g. 0.1mL+24.9mL=25mL). The total deviations of all dispensed volumes summed up are still way below the deviations of the calibrated glassware. This can improve your overall measurement uncertainty by a lot. Total cost of analysis is still mostly determent by analyst hours, not by the 1EUR pipette tip.
