My great grandmother and my grandmother both had Alzheimer’s(and likely my great great grandmother but she was never formally diagnosed). As a very young child I was very concerned my grandma would develop it, I wanted to go into some sort of research that might give me the ability to at least contribute in a small way. My grandma developed early Alzheimer’s when I was 10 or 11 and by the time I was around 20, she couldn’t remember me at all. I hope this works, not just because I worry about my own mother but for all those people who could avoid the suffering of Alzheimer’s or watching a loved one go through it. Was very disappointed that the study was poorly conducted, wouldn’t have settled the debate over amyloid but would have been good nonetheless to have a reliable datapoint

Yes it clears the analogue plaques, but it has been shown that clearing plaques alone doesn’t slow the loss of cognitive function. It’s a step in the right direction, but not a cure-or even a treatment.

It may not even be a step in the right direction, i.e. its based on the assumption that the plaques cause the disease or at least make the symptoms worse but there is now a lot of skepticism of those assumptions. If those assumptions are wrong, this isn't really going in the right direction.

Really glad we are working toward Alzheimer’s treatment. Upsetting that the US healthcare system is set up where price is inflated, everyone wants to gain, and people are left unable to obtain critical care. Glad that this market has been stimulated. Rooting for more research in this field!

I recommend dr Paul Manson finding into Treatments and how it correlate to metabolic illnesses and ways of points out ways of reversing it.

Most important part that was missed here and why people are doubting it has to do with drug attaching cells that we use to identify alzheimer's severity which might not be the cause of said illness. Meaning not only drug could be ineffective it could make detecting alzheimer's progresion more difficult.

This is a very important point. It's like cleaning up trash on the street without actually doing anything about the source of the trash. The drug only targets the biomarkers that appear in alzheimers patients, not the actual source of the disease itself. We don't know if the plaques cause the disease or if they are a byproduct of it but the ineffectiveness of this drug seems to point towards them being byproducts.

The only definetively diagnosis of Alzheimers is post mortem. You can only detect those amyloid plaques during autopsy. All current diagnoses of Alzheimers disease are based on cognitive tests (memory questionnaires) and a MRI/CT that shows brain atrophy. This is the best we have but not enough to set a differential definetive diagnosis (although statistically you would be right most of the times because Alzheimer's prevalence is high)

@@NakaUwU 9 months late, but you’re kind of right, one of the main issues with treating alzheimers is that once symptoms appear, most of the brain that will be affected already has, we are a century away from regenerating neurons, so once alzheimers progresses to the stage where symptoms occur, the patient is very unfortunately, kind of screwed. That is one of the reasons, among many other factors, why so few drugs have shown potential. We have data to support that the build up of plaque and biomarkers associated with AD occurs decades before symptoms present, so what is really damaging the brain’s neurons? We don’t really know, it could be the structure of the plaques themselves, it could be the proximity of AB42( a peptide which is known to have a degree of cytotoxicity), it could simply be inflammation in the brain, it could be the nucleation of the plaques, or something totally different, we aren’t completely sure. I would hesitate to say that the fact that the drug only improved biomarkers is indicative of one root cause, since all evidence seems to suggest that AD, once symptoms occur, your brain becoming incapable of adjusting to loss of cells. AD is a uniquely difficult disease to treat, and much of my hope for the future is that we can create a vaccine of sort that prevents the loss of brain matter before it’s too late.

@@musomike Thoughts and prayers are with you. Alzheimer's is a uniquely evil disease, and likely has the most complicated pathology of just about any disease. The good news is, however, that there is finally starting to be hope. In the past few years we have seen the first ever Alzheimer's treatments, which is remarkable considering we have known about Alzheimers for half a century; the tide may finally be turning against this despicable disease. Again, I am sorry, nobody should have to deal with Alzheimer's.

My grandmother has Alzheimer's and she recently travelled overseas when she comes back will be tough

They could just restart the trial, its was their own fault for stopping it because they thought it wasn't working. They don't get to say but people are dying. Its their own fault those people are dying now, the company should not be able to make a single cent off the drug on it until they complete the trial

i think in the clinical trails, only 27% was effective and in some patients caused brain bleeds and swelling

Before spending 56 K on a year treatment, why not RCT taladalafil sufficiently to determine if it can both reduce AD occurrence with a daily use (some research suggesting up to 69%), AND perhaps have utility and safety in improving mitochondrial function with O2 supply increase, which could help ATP production and autophagy? No RCTs because no massive profit in that?

How about RCT near infra red light stimulation to increase ROS uptake in mitochondria in brain neurons, to aid glutathione in cleaning up waste products of ATP production, and brain metabolic efficiency??? Let's see the RCTs.IMHO

If you were diagnosed with Alzheimers and had investments, would you rather spend $56k on a drug that slows down or stops the progression of your Alzheimers. Or Go into a care facility where staff have to do everything for you, such as shower you, feed you, ensure you don't wander off down the street confused and hallucinating, at a cost of 50k to 100k ? It costs my mother $90,000+ per year for her dementia care (its been going on like that since she was 60 she is now 72), and every visit her I have with her, there are more and more reduced abilities or a new set of negative symptoms. Eg, confusion, wandering, anxiety, loss of ability to put a fork in her mouth, walk, talk. 10 years on, she just sits in a chair, grinding her teeth due to the anxiety. They give anxiety drugs to stop that, but they make her fall asleep. I know the drug sounds expensive, but everything about Alzheimers is expensive.

Drugs that get accelerated approval and that are given Breakthrough Therapy Design indications often have data that isn't entirely robust. This isn't anything new at all. If you take a look at orphaned therapies, the data isn't even as robust as Aduhelm's, and the patient cost per year is even higher (300,000 USD+/patient). When it comes to these cutting edge therapies, unfortunately there is a bit of a cost between determining patient benefit, and whether or not it's even worthwhile for the pharmaceutical companies developing these drugs to even pursue, which leads to high cost.

It's not a cure. That would make no money for $corrupt$ industry. Here is cure for free: eliminate all sources of Neurotoxic Aluminum uptake. That is it: The $FDA$ allows not listing Neurotoxic Aluminum on food labels! Diet and Lifestyle factors are very small parts of this disease. Most likely, it is due to viral infections that get into the brain and hide within neurons. The Epstein-Barr virus or even HPV are thought to be a potential cause. Lots of strange new viruses these days that can cause this stuff

You understand that these are for-profit organizations? Thats how capitalism works, no company does anything for free unless theres some benefit to it. Decades of research, trials, testing, all of this costs hundreds of millions of dollars to do. People dont work for those companies for free and the company has to pay them.
