You can see Jolene fighting for a second to stay in character before giving up knowing the take is ruined.

Jolene is just reacting to a tense meeting with Dolly Parton before the shoot. Seems Dolly thought Jolene was moving in on her man…Jolene had no clue what she was talking about and found the encounter ridiculous.

I really love Jolene’s facial expressions in her bloopers. It’s crazy that some people with the best senses of humor are often cast to play non-emotional characters, like Jolene Blalock here and Brent Spiner in TNG. That guy is absolutely hilarious.

It's been said by a few character actors that comedians make the best straight men/deadpan actors because they know how they're suppose to play off them/react.

This is not a mistake. A Vulcan observing the ancient tradition of Vanu taFar, meaning to chime a bell when they enter a new orbit and being allowed to release bottled up emotions during the duration of the sound, is very much canon.
