Thank you for the video. However the explanation about the selectable marker is not quite tight. I am referring to the section from minute 11:00 to minute 13:42. I might have misunderstood you. The selection of clone having the recombinant plasmid cannot be explained here. The bigger use for Selectable Marker region is for a different issue: the transformation might not be 100% efficient and bacteria might or might not have the recombinant vector. Before transformation, you have to separate this mixture to obtain a solution that contains only the recombinant vector, which is tranformed to the bacteria (if you have a mixture of both self-annealed vector and recombinant vector to be transformed to bacteria, every bacteria that has either vector will grow in the antibiotic). Whichever bacteria does not have the vector will not grow, whichever bacteria does have the vector will grow, and of course it will have our interested gene. Just something for clarification!

Yeah, unfortunately Somu got this part wrong. I actually was watching this video just to learn about ways to prevent self-ligation of the vector. So far, I know two ways to prevent it. 1) In the ligation reaction, increase the concentration of the insert compared to the vector, like 6:1 or even higher. This increases the propability of vectors ligating with the insert. However, self-ligation is still possible. 2) Treat your vector before the ligation with Alkaline phosphatase. This enzyme will phosphorylate the ends of the (already digested) vector. Phosphate groups are negatively charged and repell each other, thus preventing contact of the vector ends with each other. In any way, to identify if the insert is correctly integrated, an analytic digest is necessary (using restriction enzymes upstream and downstream of the insertion site).
