Target proteins in a sample are detected by incubating the membrane with an antibody that specifically recognizes the protein of interest (called primary antibody). This antibody-target protein interaction is then detected either by conjugating the primary antibody with a detectable label, such as an enzyme conjugate, or by incubating the membrane with a labeled secondary antibody that only recognizes the primary antibody.
