These lectures describe the principles of dark field and phase contrast microscopy, two ways of generating contrast in a specimen which may be hard to see by bright field. The lecture describes how the phase rings work to generate interference between the diffracted and undiffracted light.

Many thanks for this excellent information. I have an Olympus BX50 and have never known the complexities of the light delivery in my microscope... until today.
8:40 says NA for condenser is less than objective, but NA_cond > NA_obj
@30:00 that is brightfield not darkfield. Was looking for Darkfield phase contrast content.

the back ground slide at 16:30 doesnt make sense. If the diffracted light is focused on the quarter wave plate, then how can it be focused too on the image plane ? Moreover if the planar/collimated illumination focuses on the image plane, meaning the focale distance is the image plane distance, then how is it that the specimen is focused at the same point ??!! The specimen could not have an image if it's closer to the lens than the focal distance, and if it's further away then the image must be further away than the focal distance....
I didn't get it. what exactly was done/happened to the phase of the undiffracted light and how was it different in the earlier method without the circular disc ? Also, what purpose does converting the light rays into annular cones serve ?

is there any way to see lipids stained with a fluorescent dye Nile Red under phase contrast microscope? Plz tell if anyone knows because I don't have fluorescent microscope
