Great video! Will fixatives affect the staining of nuclei by Hematoxylin (in terms of the bouin's and paraformaldehyde)?

Hi Qiran, yes, fixation technique can impact significantly on staining outcomes. The picric acid in Bouin’s fixation can improve retention of trichrome stains making them seem more vibrant. I haven’t examined specifically affects on nuclear staining, but pH can certainly affect binding and retention of hematoxylin.
