In tobacco for callus formation, small sections are taken to avoid any fungal or bacterial growth. The ratio of auxin and cytokinin are responsible for the formation of organ formation. For the development of roots concentration of auxin should be higher and for the formation of shoots, cytokinin concentration should be higher. Both acts oppositely to each other and helps in the formation of the organ.
