mantissa is the value after the decimal point of binary no. you have calculated (after shifting the decimal point to left) [8:50], and 754 is a standard name given by IEEE that you are converting you number into

1 and 0 zeros can only be represented in binary. Like 0.5 in decimal = 0.1 in binary. And 0.25 in decimal = 0.01 in binary.
