How to Make and Use a Still Air Box

This video is about the process of making and using a still air box for growing mushrooms. It is made from a large plastic container. Two armholes are melted into the container with a coffee can to make perfect holes. The edges of the holes are made smooth with some sandpaper. The second half of the video explains some good practices for using a still air box to increase your chances of success by reducing the risk of contamination.

    Don't wipe the freshly flamed needle/scalpel with anything. Defeats the purpose..

    Do not flame sterilize inside the SAB!!! First, you're putting yourself in danger of igniting the isopropyl fumes, second the flame produces a lot of circulation inside the still air box, defeating the purpose of STILL AIR

    Why flip the tub for when you use the gloves setup? Why not keep the lid on the bottom, same as without the gloves?

    So question, I’m over thinking it but the sterilizing the box thing before aren’t you letting in more contam opening it an putting everything inside? I had a bad lc from the jump so made me feel like I messed everything up an I went crazy with iso an sterilizing. Waiting for my new lc now an redoing this

    Thanks for the video and a clip of what it looks like working in the box. Wondering - is it necessary to have a grill on the bottom so contaminants "fall through the cracks" below, and not on the same surface where your working items are? And for SABs, is 'bigger better'? My guess is that the larger the box, the less likely air movement will be an issue (due to hands/arms moving around the hole?) One last one - I see people complain about seeing through the plastic. But I don't see much online about people building SAB's out of glass (ie using old window glass, or even fish tanks). Any reason why someone wouldn't build a SAB out of glass (with silicone glue/sealant). Thanks!

    Just more labour intensive. It's easier to spend the same ammount of time building a laminar flow hood.

    A glass SAB would be nice. It will disinfect easier too.

    Nice job, I like the idea of bleach instead of alcohol , you said 10% bleach but most consumer bleach is only 6% available chlorine ( usually hypochlorite ). Did you find a stronger bleach somewhere or did you mean you diluted household bleach at a 9:1 ratio with water ? I have hypochlorite available to me , its twice as strong as household bleach ( usually 12% but like bleach loses strength over time ) the weaker the solution the longer it takes to disinfect. I'm thinking a .3 or .4% chlorine solution would be plenty good. The smell is a bit worse than alcohol but worth it to eliminate the torch issue...cheers

    Fwiw, I just picked up a 90 qt Room Essentials bin from Target for 13 bucks. It's maybe 10-15% larger than OP's. Flat lid, no handles/latches, and it sits flat upside down.

    Glove boxes are no longer recommended as they have a tendency to create more air movement than a basic still air box. They are old tek that has been deprecated for several years now. Also, never flame anything inside a still air box.

    The flame is what I was here to find out about. If you spray the entire area, aerosoloizing 70% isopropyl alcohol, how is that not risking combustion? Do you flame sterilize outside the box and then move the sterile tool inside?

    @@benson733 Thank you. There's so many more steps and precautions to take with a SAB than with a laminar flow hood, but not everyone has $1000-$2000 to spend on a hobby 😒

    Is this just for DIY stuff? That’s interesting I was under the impression real glove boxes (used in university labs) are basically completely sterile environments. I can definitely see how a DIY glove box might force more air movement tho, I’ve never used one

    @@big_hat_logan I mean I use it for mycology and diy. I'm thinking the term glove box refers to a different type of box with gloves attached and that creates much more air flow than a standard still air box with only holes cut. You disinfect and then spray the box full of bleach water or even soapy water and let it sit for 10+ minutes. Then just use proper sterile techniques and you should have good low contamination rates. Laminar flow hoods are king though.

    I'll just add that bleach + rubbing alcohol = chloroform. Use one or the other in your workspace, never both.
