Hi Dr. Gary! I have difficulty viewing the allium cepa root tips. I cant see clearly the cells of the root. Can you you please give me some techniques to view it clearly .Hoping for faster reply thank you

What happens if you using oil immersion on 100x and accidentally go toward the labeling ?

Why not first focus with the 40x, move to the intermediary position between the 40x and the 100x, place a drop of oil on the slide, and then move to the 100x?

Hi! i'd like to ask, isn't it necessary to use scanner, low, high then oil immersion objective? or is it alright to just skip all 3 then to oil immersion?

When looking at objects that can be seen when using a 10X objective (100X) starting at low power and rotating the objective lenses to obtain higher magnification works well. When viewing bacteria, most are too small to be seen when using a 10 X objective. In this case it is faster to start right out with the 100X oil immersion objective (1000X) as described in my video.

Finnaly got my microscope.But i was looking at a blood puddle and my microscope couldnt focus on the puddle but kept going out of focus.Im pretty sure this is beacuse i have old plastic top cover for the slide and its not a problem on my microscope but if it is i need help.

I use motor oil. Works great.

This is a very dangerous method that can ruin both the objective and the specimen. Long before 100x objective is used, coarse knob is not turned at all. Coarse adjustment knob is used with 4x and 10x objectives, then a 40x objective is set and image is focused with a fine adjustment knob. Revolver is set between 40x and 100x, immersion liquid is added on the slide and 100x objective is lowered into the drop and rocked a bit to avoid air bubbles and ensure complete wetting. Then the fine focus adjusting is a matter of tiny turn because 40x and 100x under oil have nearly the same focuses. 100x is so close to the specimen (around 100 micrometres) that moving the coarse adjustment is a good way to press the objective into the slide which often results in damage. Your method requires lots of turning of the fine knob which is detrimental to the fine adjusting system. It loses rigidity that way. I've seen it lots of times with students.

You also need to put oil between the condenser and the slide.

Only on oil immersion condensers. Most condensers are not oil immersion and immersion oil may cause damage.
