What happens in dota2 when a player loses connection? Is there also an AI or do you go on as 4? Also, will the loss count then?

Depends on the situation. When someone disconnected, that individual will get a 5 minutes countdown. If the player failed to reconnect back to the game on time, he will be treated as a leaver and will receive leaver penalties. Other players can pause the game to stop the countdown, and gain control of the disconnected player's hero after the countdown is over. Other players will also gain additional golds when someone completely leave the match. Even if the player reconnected afterwards, as long as the 5 minutes timer is out, he will still be treated as a leaver and his penalties remains. If the game is an unranked match, then once the timer is out the game will not be counted and everyone is safe to leave without receiving leaver penalties. If the game is a ranked match, and if someone disconnected and leave before the first blood or within the first 5 minutes of the game, then the game will also not be counted, and everyone is safe to leave without penalties. If, in a ranked match, some disconnected and leave after first blood or pass the first 5 minutes, then the match however will be counted. Leavers in a ranked match will always receive a loss record despite the results of the match.
