In this video, I have explained how to calculate the value of Km and Vmax for an enzyme-substrate reaction using the Michaelis-Menten equation. We will use Excel, a common data analysis software to calculate Vmax, Km, Kcat. Hopefully, you will like this video. In excel we used the line plot, simple statistics and solver to get the actual Km, Vmax and Kcat value of enzyme kinetics.

    To calculate enzyme concentration [E] in Kcat calculation. Kcat = Vmax/[E] Example: Vmax = 200 umol/mg/min Enzyme concentration = the amount of the enzyme used in the assay or reaction from Vmax - its 1 mg. so what is the concentration of enzyme [E] in 1 mg (amount of enzyme/mg) lets say its molecular weight is 28KDa = 28000 Da = 28000 gms/mole = 28000 mg/mmol = 28mg/umol (1 mole = 1000 mmol, 1 mmol = 1000 umol) 1 mg of enzyme = 1/28 umol = 0.0357 umol so concentration of enzyme [E] = 0.0357 umol/mg Kcat = Vmax/[E] =200 umol.mg-1.min-1/0.0357 umol.mg-1 Kcat = 5602 min -1
