    Hello I have a question, after ds cut using cas9, how new fragments/ nucleotides are introduced into that cut. Are they introduced in primers of flanking region or through target sequence. Pl. elaborate

    Actually the DNA breaks are repaired using a method used by cells called NHEJ (Non homologous end joining) where the broken ends are directly ligated without the need of homologous template.

    @@NehasBiology thank you for this information but what about HDR pathway, where we are providing the flanking homologous regions of target sequence.

    HDR is also seen in CRISPR-Cas9. And here one strand of homologous DNA duplex results in the formation of hybrid DNA which can then help in DNA repair process.
