Thanks for another great video! When you switched between brightfield and phase contrast at 03:50 did you use a different objective, or can phase contrast objectives also be used for brightfield?

I just rotated the condenser, same objective. Phase objectives can be used for bright field as well. There is a small decrease in image quality, however. In the next video I will talk exactly about this topic and show you comparison images.

When using phase contrast, do you have to stain the specimen before you look at them? And also, what do organ cells look like under phase contrast??

The point of phase contrast is that it is not necessary to stain cells (which could harm them) and still be able to see certain details.

How does Phase contrast compare to polarizing filters?

Looks like you don't have a filter holder under that phase condenser, is that right? So maybe it might not be the best condenser to use when doing brightfield, at least if you are doing rheinberg or oblique illumination.

I'm new in the microscopy world, I've been watching your videos and your love for this field encouraged me to start this. I'm interested in observing plant cells and plant tissue. What microscope would you recommend me? My budget is less than $400, do you think I can get something good with that? What's your opinion? Thank you 😊

I generally recommend the Swift SW380T (I made review videos) or the Omano OM36 (also a video available), if you live in the US. For Phase contrast, you have to spend much more than $400 and I would not do that right now. Many things to see using normal brightfield.

You left out WHY the other phase contrast condenser did not work! I bought a very nice Olympus phase contrast condenser (originally expensive metal version!) from ebay and am not getting too good of results. (can't see bacteria like you show in the video) How do I pick which phase contrast objectives to buy? (I have a model EH scope) The Olympus phase contrast objectives are expensive even on ebay! Can I use some of the Amscope cheap phase contrast objectives?

I mentioned this in another video: kzread.info/dash/bejne/eWV3mqeDicupdpc.html The problem is that my old condenser worked with the old Olympus objectives, but the newer objectives not. Reason being, that the size of the objective phase ring and condenser phase annulus did not match in size. The new condenser now matches the new objective. The lesson that I learned, is that objectives and condenser should match. The person who sold me the condenser told me that Euromex phase condensers for the Novex B seem to be compatible to Olympus regarding ring size and condenser mount. At least he was able to get the 40x to work. So maybe some modern manufacturers simply copy traditional systems, instead of defining their own standard? I don't know for certain. I guess you would be able to use the Amscope objectives (if they have RMS threading, not infinity) then they should fit. But then the condenser must match as well. Expect to possibly lose parfocality with the other objectives (ie refocusing necessary when changing objectives).

Hi, Do you have model numbers of condenser and objectives for Olympus bx51 and where to buy the matching sets ? What magnification levels are best used for PC? I use 20, 40x for DF for live blood. Thank you

The phase ring in the objective and the phase annulus in the condenser must have the same size. They must overlap.

I have a neogenesis systems trinocular brifhtfield/darkfield microscope. Can i convert it to a phase contrast microscope? Am i required to invest in special objective?

Oliver can u make a video on DIC microscopy and explain how does it work and in which cases it is used plsss hope u dont mind

40x is enough. 100x needs oil immersion, which is a disadvantage.

Hi Oliver, informative video as always. You now have both Olympus short barrel (in your CHA) and long barrel (in your CH40) phase contrast PL objectives. Are the newer long barrel ones better than the older short barrel? I have both a BH2 and CHA so I also have the flexibility to purchase either, with the correct condenser of course. Thanks.

If I have phase contrast objectives and use a brightfield condenser, will I be seeing pure brightfield images or do you need a set of regular non phase contrast lenses to see in pure brightfield?

Phase Contrast Objectives with brightfield condenser will produce a regular brightfield image. The image quality is a bit lower, but not much.

If you ever consider making a video comparison on your Brightfield condenser with Brightfield Objectives vs your Phase contrast condenser setup (non-brightfield illumination) with Phase Contrast objectives that would be awesome! Pretty much pure phase contrast vs pure brightfield. Thanks again!

for a phase contrast microscope, indeed not very expensive.

instead of bying PH u should buy 2 wollastone prisms

you make darkfield filters, why not phasekontrast filters

Darkfield gives it like a 3 dimensional look

on my opinion for an amateur dark field is better than phase kontrast and its much cheaper
